幽门螺杆菌对胃上皮细胞株GES-1凋亡及XIAP表达的影响

目的观察幽门螺杆菌（Hp）慢性感染对胃上皮细胞凋亡及X凋亡抑制蛋白（XIAP）表达的影响。方法通过Hp SS1与人非肿瘤性胃上皮细胞株GES-1共培养，建立GES-1模型细胞，检测模型细胞凋亡率及胃上皮细胞XIAP mRNA的表达。结果Hp感染8周、12周、16周模型细胞凋亡率分别为64．55％、60．58％、45．52％，模型胃上皮细胞的XIAP mRNA实时定量PCR的CT值分别为0．901、0．842、0．796，明显低于对照组细胞凋亡率75．52％（P〈0．05）及XIAP mRNA的CT值1．030（P〈0．01）。结论Hp慢性感染能抑制胃上皮细胞凋亡，其机制可能与上调XIAP mRNA表达有关。

Effect of chronic helicobacter pylori infection on apoptosis and expression of X inhibitor of apoptosis protein in gastric epithelial cells

Objective: To establish a cell model by chronically infecting human gastric epithelial cells with helicobacter pylori(Hp),and to investigate the effect of chronic Hp infection on apoptosis and expression of X inhibitor of apoptosis protein(XIAP) in gastric epithelial cells.Methods: Human non-tumor gastric epithelial cells GES-1 were cocultured with an Hp strain SS1,in order to obtain GES-1 model cells.Apoptosis of the model cells were measured by Annexin V/PI staining and XIAP mRNA expression detected by real-time quantitative PCR.Results: The apoptotic rates of model cells which Hp infected 8 weeks,12 weeks and 16 weeks(64.55%,60.58%,45.52%) were significantly lower than those of control cells(75.52%)(P< 0.05);The CT value of the model cells which Hp infected 8 weeks,12 weeks and 16 weeks of XIAP mRNA expression detected by real-time quantitative PCR were lower than that of control cells(P<0.01).Conclusion: Chronic Hp infection induces apoptosis resistance in gastric epithelial cells which is possibly by the mechanism of up-regulating XIAP mRNA expression.
