摘    要:研究切割bcl-2mRNA核酶诱导胆管财亡的形态学改变。方法将合成的切割bcl-2mRNA核酶民真核细胞表达载体重组,并用重组后的载体转染人胆管癌细胞,通过光镜及扫描和透射电镜手段,观察转染后的胆管癌细胞,通过光镜及扫描和透射电镜手段,观察传染后的胆管癌细胞凋亡的形态学改变。

Abstract:AIM To study the morphologic transform of apoptosis in human cholangiocarcinoma cells induced by bcl 2 mRNA cleaving ribozyme (RZ). METHODS Synthesized gene of bcl 2 mRNA cleaving RZ was recomposed with a retrovirus carrier that can transcribe and express in eukaryotic cells and was transfected into human cholangiocarcinoma cells. The morphologic transform of apoptosis in human cholangiocarcinoma cells was observed by light microscope as well as scanning electronic microscope and transmission electronic microscope. RESULTS Typical morphologic transform of apoptosis appeared in transfected cholangiocarcinoma cells. CONCLUSION The bcl 2 mRNA cleaving RZ can cut down bcl 2 expression level of cholangiocarcinoma cells remarkably and induce the cell apoptosis spontaneously.
