弓形虫感染家兔血液中虫体的动态观察

摘    要:目的探明虫体在家兔血液中动态变化情况。方法用RH株弓形虫速殖子105个/只的剂量腹腔感染雄性家兔16只。在家兔感染前后分别采集家兔血液,抗凝处理后,-40℃冻存备用,用实时定量PCR检测弓形虫感染家兔血液中弓形虫DNA,依据对照计算虫体拷贝量,绘制感染后时间与血液中虫体含量的动态曲线图。结果雄兔弓形虫感染后4d血液中即可检测到虫体DNA,血液中虫体含量为217.887×103个/ml,血液中虫体含量在感染后8 d达到高峰期,血液中虫体含量为248.019×103个/ml,然后虫体含量逐渐下降,在感染后121 d雄兔血液中虫体密度为1.45×103/ml。结论弓形虫感染雄兔血液中虫体含量随感染后时间不同而变化。

Abstract:Sixteen rabbits were infected with Toxoplasma gondii intraperitoneally with 10~(5) tachyzoites of the RH strain,and the blood samples were collected before and after infection.Quantitative real time PCR was used for the dentection of T.gondii DNA in blood samples,and the densities of T.gondii in blood were calculated in reference to the known DNA of T.gondii.The figures of the kinesis in density of T.gondii in blood from infected rabbits were drawn with Microsoft Excel.It was found that the DNAs of T.gondii in blood of the infected rabbits could be detected around 4 days after infection.During that time,the density of T.gondii in blood of the infected rabbits was 217.887 x 10~(3) /ml,it rose up to its peak value of 248.019 x 10~(3)/ml at 8 th day after infection and then it gradually declined.However,it remained to be detectable in blood of the infected rabbits up to 210 days after infection with a density of 1.43 x 10~(3)/ml..It is evident that the densities of T.gondii in blood of the infected rabbits shows changes in pace with time of infection.
