摘    要:目的:观察模拟咬合压力对人牙周膜成纤维细胞环氧化酶-2(COX-2)基因表达的影响。方法:原代培养人牙周膜成纤维细胞,应用可控压力细胞加载装置模拟咬合压力,分别施加30、60、90kPa的间歇性压力,每次15min,每天3次,在3、5d后提取细胞总RNA,定量后点于尼龙膜上,同时利用引物扩增COX-2基因的一段特异性片段作为cDNA探针,将探针同位素标记后与膜杂交,以观察COX-2在mRNA水平表达的变化。结果:加载模拟咬合压力的人牙周膜成纤维细胞COX-2表达水平明显高于对照组,而且在一定范围内表达水平与加压力值成剂量依赖关系;各加压组加压3d与5d之间细胞COX-2mRNA表达无明显差异。结论:加载模拟咬合压力条件下培养的人牙周膜成纤维细胞COX-2基因表达增高。

The influence of simulated occlusion pressure on the expression of cyclooxygenase-2 in human periodontal ligament fibroblasts
Abstract:Objective: To investigate the influence of simulated oc clusion pressure on the cyclooxygenase-2 (COX-2) gene expression in human peri odontal ligament fibroblasts (HPLFs). Methods: An adjustable pressure-loading a pparatus was used to load intermittent simulated occlusio n pressure(kPa) at 30, 60 and 90 respectively On primarily cultured HPLFs for 3 or 5 days, 15 min for each time and 3 times a day. Cell general RNA was extrac ted; COX-2 cDNA probe was prepared by RT-PCR and labeled with isotope, CO X-2mRNA expression was examined with dot blotting hybridization. Res ults: COX-2 expression level in HPLFs loaded with intermittent sim ulated occlusion pressure was significantly higher than that in the controls in a magnitude-dependent manner. But there was no difference between the result s of 3-day-treatment and 5-day-treatment in each pressure group. Conclusion: COX-2 gene expression increases under intermittent sim ulated occlusion pressure.
