山东荣成八代按蚊分类地位的探讨

摘    要:目的探讨山东荣成八代按蚊的分类地位。方法于2009年8月底在山东荣成西郊王家村采集饱血八代按蚊,饲养得子代,对子代成蚊进行形态学鉴定。抽提各型成蚊足的DNA,PCR扩增rDNA-ITS2片段后测序,并用DNAstar7.1软件对所获荣成八代按蚊的rDNA-ITS2序列与文献记录的山东荣成八代按蚊(YSD,AY306128)、四川省八代按蚊(YSC,AY170925)、辽宁省八代按蚊(YLN,AY170923)和韩国八代按蚊(YK,AF146749)的相关序列进行多重比对。结果共采集饱血八代按蚊56只,其中7只顺利产卵,饲养共获得子代成蚊354只,包括八代按蚊型(Y型,成蚊翅脉端白斑和V5.2顶繸白斑均有)240只、近黑按蚊型(P型,两者均无)8只和杂合型(H型,两者其一)106只。亲本和子代成蚊各型间rDNA-ITS2段序列(JN865249,JN865246,JN865247,JN865248)同源性为98.7~100%,与四川省八代按蚊、辽宁省八代按蚊和韩国八代按蚊的rDNA-ITS2段序列同源性分别为98.5%~99.3%、98.7%~99.6%和98.7%~100%,而与原山东荣成八代按蚊同源性仅为6...
