急性坏死性胰腺炎模型大鼠血清TNF-α/IL-10的动态变化及丹参的干预效应研究

摘    要:目的探讨TNF-α/IL-10在急性坏死性胰腺炎(ANP)模型大鼠血清中的动态变化及丹参的干预效应。方法取SD大鼠96只,造模后随机分为胰腺炎(P)组、丹参治疗(T)组,并另设对照组(C)组,每组32只,测定其血清TNF-α、IL-10水平及TNF-α/IL-10变化,同时取胰腺组织进行病理评分。结果与C组比较,P组血清TNF-α、IL-10水平明显升高(P<0.05或P<0.01),病理改变明显加重;与P组比较,T组各时相血清TNF-α、IL-10水平明显降低(P<0.01或P<0.05),病理改变明显减轻。结论ANP发生时TNF-α/IL-10比例失调,呈现由升高-相对平衡-降低的模式;丹参可下调TNF-α/IL-10比值,其机制可能与其双向调节TNF-α、IL-10水平有关。

Abstract:OBJECTIVE:To study the dynamic changes of TNF-α/IL-10in model rats with acute necrotizing pancreatitis (ANP)and to study the intervention outcome of salvia miltiorrhiza.METHODS:A total of96rats were randomly divided into three groups(each with32rats):pancreatitis(P)group,salvia miltiorrhiza treatment(T)group and control group(C),the blood samples of rats in each group were taken to determine levels of TNF-αand IL-10and the ratio changes of TNF-α/IL-10.Meanwhile,pancreas tissue sample was collected for pathological scoring.RESULTS:Group P had significantly higher serum levels of TNF-αand IL-10than did group C(P<0.05or P<0.01)and the pathological changes in this group were significantly aggravated.As compared to group P,levels of serum TNF-αand IL-10in T group were significantly lowered(P<0.01or P<0.05)and the pathological changes were significantly alleviated.CONCLUSION:When ANP occurred,the TNF-α/IL-10was in a disproportionate model:from the initial increase to relative equilibrium then to reduction.Salvia miltiorrhiza can lower the ratio of TNF-α/IL-10,the possible mechanism is that it can perform a two-ways regulation on TNF-αand IL-10.
