亚慢性甲苯染毒对大鼠卵巢的毒性作用

摘    要:目的:探讨亚慢性甲苯染毒对大鼠卵巢的直接毒性作用。方法:将40只雌性大鼠随机分为对照组和甲苯染毒组。对照组给予1 ml不含甲苯的玉米油,染毒组甲苯剂量从低到高分别为35、70、330 mg/kg,连续染毒40~45天,观察甲苯对卵巢脏器系数、组织结构的影响;计数各级卵泡及黄体数目,用TUNEL法检测颗粒细胞凋亡率,并测血清E2、P的浓度。结果:①中、高剂量甲苯组卵巢脏器系数较对照组降低,差异有统计学意义(P<0.05)。②4组之间原始卵泡数目无显著改变,随着染毒剂量的增加,生长卵泡减少,闭锁卵泡增加,黄体数目减少,且中、高剂量组与对照组相比,差异有统计学意义(P<0.05)。③中、高剂量甲苯组颗粒细胞凋亡率(26.6%,35.3%)高于对照组(10.9%),差异有统计学意义(P<0.001)。④中、高剂量甲苯组E2、P较对照组降低,差异有统计学意义(P<0.01)。结论:亚慢性甲苯染毒对大鼠卵巢有毒性作用,且与甲苯剂量呈正相关。

Abstract:Objective:To explore the direct effects of subchronic exposure to toluene on ovaries of female rats.Methods:40 female rats were divided into control group and toluene group.Rats in control group were administrated 1 ml maize oil without toluene,while rats in toluene group were administrated toluene for 40~45 days,the concentrations were 35,70,330 mg/kg,respectively.The effects of toluene on organ coefficients of ovary and organ structure were observed;the amounts of follicles at different levels and corpus luteum were calculated;the apoptosis rate of granulosa cells was detected by TUNEL method;the concentrations of estrogen and progestogen in serum were detected.Results:①The organ coefficients of ovary in middle and high dose toluene groups were lower than that in control group(P<0.05).②There was no significant difference in amount of primordial follicles between the four groups,the amounts of developing follicles and corpus luteum decreased,while the amount of atretic follicles increased with concentrations of toluene,there was significant difference between middle and high dose toluene groups and control group(P<0.05).③The apoptosis rates of granulosa cells in middle and high dose toluene groups(26.6%,35.3%) were higher than that in control group(10.9%),there was significant difference(P<0.001).④The levels of estrogen and progestogen in middle and high dose toluene groups were lower than that in control group(P<0.01).Conclusion:Subchronic exposure to toluene has an venenatus effect on ovaries of female rats,which is related to the dose of toluene.
