| 摘 要： | 目的:从粗毛褐孔菌中分离麦角甾醇并建立测定含量的HPLC法。方法:采用硅胶柱层析法及重结晶法分离纯化,波谱分析鉴定其结构;高效液相法测定其含量,用十八烷基硅烷键合硅胶色谱柱(KromosilTMC185μm,150mm×4.6mm),流动相为甲醇-水(90∶10),流速1.0mL/min,柱温室温,检测波长270nm。结果:麦角甾醇在44~220μg/mL之间与峰面积呈良好的线性关系,r=0.9998(n=5);平均回收率为99.71%,RSD为2.62%(n=6)。结论:该方法简单准确为粗毛褐孔菌药材质量评价提供了可靠依据。

| Abstract: | Objective:To abstract ergosterol from Xanthochrous hispidus(Bull.exfr.)Pat.and determinate it's contains by HPLC.Methods:The content of ergosterol was detected by HPLC,Chromatographic separation was carried out with an KromosilTM C18(5μm,150mm×4.6mm) under isocratic conditions methanol and water(90∶10) as the mobile phase at a flow-rate of 1.0mL/min.The UV detection wavelength was set at 270nm.Results:Ergosterol has a good linearity within 44～220μg/mL r=0.9998(n=5) and the average recovery was 99.71% with RSD 2.62%.Conclusions:This method is simple,accurate and can be used for the quality control of Xanthochrous hispidus(Bull.exfr.)Pat. |
