哮喘模型大鼠肺组织血管内皮生长因子的表达及布地奈德干预的影响

摘    要:目的①观察血管内皮生长因子(VEGF)在哮喘模型中表达的变化;②研究糖皮质激素干预对VEGF及其mRNA表达的影响。方法 36只清洁级雄性SD大鼠随机分为对照组(C组)、哮喘组(A组)和布地奈德干预组(B组),制备大鼠哮喘急性模型。末次激发后24h,留取标本用于免疫组化和原位杂交检测。光镜观察肺组织病理变化。结果①肺组织切片HE染色结果光镜下表现:C组示支气管和血管周围未见炎性细胞浸润,细支气管平滑肌未出现增殖情况。A组可见细支气管形态不规则,平滑肌轻度肥厚;支气管壁和血管壁可见较多的炎性细胞浸润;支气管腔内见粘液栓和炎性渗出物;B组肺组织病理改变同A组相比明显减轻,但炎性细胞浸润等改变未完全消失。②免疫组化法示肺组织VEGF蛋白表达结果:A组肺血管壁及支气管壁VEGF表达的吸光度值均显著高于C组(P〈0.01),B组高于C组(均P〈0.05),A组显著高于B组(均P〈0.01)。③原位杂交法示肺组织VEGFmRNA表达结果:A组肺血管壁及支气管壁VEGFmRNA表达的吸光度值均显著高于C组(P〈0.01),B组高于C组(均P〈0.05),A组显著高于B组(均P〈0.01)。结论①模型大鼠哮喘急性发作早期VEGF的表达是增加的。ASMC、气道上皮细胞、血管平滑肌细胞是其重要的来源细胞。②糖皮质激素可抑制VEGF及其mRNA的表达。

Abstract:OBJECTIVE ①To investigate the changes of vascular endothelial growth factor(VEGF)in asthmatic rats.②To study the regulation role of glucocorticoid on expression of VEGF.METHODS The rat model of asthma was established by the ovalbumin(OVA)challenge methods.Thirty-six male rats were randomly divided into three groups on average,24 hours after the last challenge,the animals were sacrificed.lung tissue were collected.RESULTS ①The observation results of light microscope:Under light microscope,HE-stained lung tissue in group C cann't find inflammatory cells infiltration around the bronchus and the vascular,and there wasn't bronchiole smooth muscle proliferation as well.While in group A it was showed that the form of bronchial wall was irregular,bronchiole smooth muscle slightly proliferation;there were inflammatory cells infiltration around the bronchus and the vascular;bronchus mucus bolting,goblet cells proliferation;there were inflammation cells infiltration in lung interstitial and alveolar.Compared with group A,the pathological changes of lung tissue in group B significantly alleviated,but the changes(such as inflammation cells infiltration)still existed.②The results of immunohisto-chemistry:Immunohistoche-mistry showed that the optical density of VEGF expressions around lung vessel and bronchus wall in group A were significantly increased than those in group C(P0.01);and the optical density of VEGF expression around lung vessel and bronchus wall in group B were significantly increased than those in group C(P0.05);the optical density of VEGF expression around lung vessel and bronchus wall in group A were significantly higher than those of group B(P0.01).③The results of hybridization in situ:Hybridization in situ showed that the optical density of VEGF mRNA expressions around lung small artery and bronchus wall in group A were significantly increased than those in group C(P0.01);the optical density of VEGF mRNA expression around lung small artery and bronchus wall in group B were significantly increased than those in group C(P0.01);and the optical density of VEGF expression around lung small artery and bronchus wall in group A was significantly higher than those of group B(P0.01).CONCLUSION(1)The expression of VEGF is increased in asthmatic rats,and their important sources include ASMC,airway epithelial cell and vascular smooth muscle cell.(2)Glucocorticoid can significantly decrease the expressions of VEGF and VEGF mRNA.
