摘    要:目的:观察细胞松驰素B(cytochalasinB,CB)对K562细胞形态和增殖能力的影响。方法:以人红白血病K562细胞为细胞模型,经不同浓度的CB作用不同时间(4-48h),通过相差显微镜和Giemsa染色观察细胞形态学改变。细胞计数法绘制细胞不同条件下的生长曲线。结果:细胞形态发生显著变化,处理组细胞出现不同程度的脱核,细胞生长受到显著抑制。结论:CB对K562细胞的脱核作用与其浓度呈正相关,随作用时间延长而增强;秋水仙素有显著提高CB诱发细胞脱核的作用;CB对细胞的增殖能力有显著抑制作用。

Abstract:OBJECTIVE: To observe the effect of cytochalasin B on the denucleation of K562 cells. METHODS: K562 cells were grown in RPMI 1640 medium supplemented with 10% calf serum and 4 microL cytochalasin B (CB). Denucleation was induced in the cultured cells by CB, and the cells were examined by phase contrast microscopy and Giemsa staining respectively. RESULTS: The denucleation of K562 cells induced by CB was clearly observed, and the cell proliferation was obviously inhibited. CONCLUSION: The denucleation efficiency of K562 cells is positively correlated with CB concentrations and the duration of CB treatment. CB at low doses may inhibit the cell proliferation and at high doses causes cell death.
