野生蚕抗菌肽对癌细胞的杀伤作用及其超微结构的研究

摘    要:目的从野蚕蛹血淋巴中分离纯化抗菌肽,并研究野蚕抗菌肽对体外培养癌细胞的杀伤作用及超微结构的影响。方法采用MTT法检测野蚕抗菌肽对癌细胞的杀伤作用,并用透射电镜观察野生蚕抗菌肽对癌细胞超微结构的影响。结果野蚕抗菌肽对体外培养的A549、HepG2、SW620、HME1四株癌细胞具有较强的杀伤作用,而对HNE1鼻咽癌细胞和MCF-7乳腺癌细胞杀伤作用较弱,透射电镜观察发现野蚕抗菌肽处理癌细胞后其细胞表面微绒毛减少、细胞膜破损,线粒体肿胀并形成空泡,核被膜消失、核浓缩聚集靠边呈块状或新月状凋亡小体。结论野蚕抗菌肽可有效地杀伤人体不同系统的癌细胞,并诱导HepG2癌细胞凋亡。

Abstract:Objective To purify wild antibacterial peptide from the haemolymph of silkworm Bombyx mori, and to study the killing action of antibacterial peptides on cancer cells and observe their ultrastructure. Methods We assessed the feasibility of using the MTT assay as a measure of cell viability in antibacterial peptide sensitivity testing in A549, HepG2, SW620, HME1, HNE1 and MCF-7 tumor cell lines. The ultrastructure of HepG2 cells treated with the antibacterial peptides was observed by the scanning and transmission electron microscope. Results The purified antibacterial peptides killed the cancer cells cultured in vitro and brought about a series of pathological changes on cancer cells. The cancer cell membrane and nuclear enclear envelope were broken, mitochondrion was swollen and intracellular substances were leaked out, the cells died soon. Conclusion The wild antibacterial peptide can kill the cancer cells cultured from different system in vitro and induce the HePG2 tumor cell apoptosis.
