摘    要:目的:研究二甲双胍(Met)对体外培养子宫内膜癌Ishikawa细胞增殖、凋亡、迁移的影响并探讨其机制。方法:Ishikawa细胞经Met 作用后,MTT法检测细胞增殖情况;流式细胞法检测细胞凋亡及周期分布;划痕法和Transwell法检测细胞迁移及侵袭能力;蛋白质免疫印迹法检测胰岛素生长因子-1(IGF-1)的表达水平。结果:经Met处理后,Ishikawa细胞增殖、迁移和侵袭能力受到明显抑制,凋亡率显著增高;随着Met浓度地升高,IGF-1蛋白表达水平显著下降(P<0.05,P<0.01)。结论:Met可通过下调IGF-1蛋白的表达对子宫内膜癌Ishikawa细胞产生抑制增殖、诱导凋亡和降低迁移及侵袭等作用。

Abstract:Objective:To study the effects of metformin(Met) on proliferation,apoptosis and invasion of human endometrial carcinoma Ishikawa cells in vitro and explore its mechanism.Methods:The growth and proliferation of Ishikawa cell were detected by MTT method.Cell apoptosis and cell cycle were detected by the flow cytometry.Cell transfer and invasive ability was measured with cell scratch assay and Transwell method.The expression of IGF-1 was detected by Western blot.Results:Met could inhibit cell proliferation,induce cell apoptosis,decrease S phase cells and decrease transfer and invasive ability of Ishikawa cells in a dose-dependent manner and the level of IGF-1 was markedly suppressed by Met(P<0.05,P<0.01).Conclusion:Met suppressed the proliferation,induced apoptosis and decreased migration and invasion in Ishikawa cells by down-regulating the expression of IGF-1.
