人肝癌组织中S1P1和AT1R蛋白及mRNA表达的初步研究

目的:检测S1P1和AT1R蛋白及mRNA在人肝癌组织中的表达。方法:对18例人正常肝组织和23例人肝癌组织进行HE染色。采用免疫组化、Real-time PCR及Western blot方法检测人正常肝组织和人肝癌组织中S1P1及AT1R蛋白及mRNA表达。结果:S1P1及AT1R在人正常肝组织的细胞膜和细胞浆中可见弱的棕黄色颗粒沉着，肝癌组织的细胞膜和细胞浆中可见强的棕黄色颗粒沉着。与正常肝组织相比，肝癌组织中S1P1和AT1R蛋白及mRNA表达水平显著增高(P<0.05)，两者呈现一致趋势。结论:S1P1及AT1R在人肝癌组织中的蛋白及mRNA表达水平呈一致升高的趋势，提示两种分子之间可能存在一定的相关性，为进一步研究其在肝癌发生发展中的关系提供了一丝新线索，也为临床上肝癌的诊断和防治提供一些新策略。

Preliminary study on protein and mRNA expressions of S1P1 and AT1R in human liver cancer tissues

Objective:To investigate protein and mRNA expressions of S1P1 and AT1R in human liver cancer tissues.Methods:HE staining of 18 normal liver tissues and 23 human liver cancer tissues was observed.Moreover，protein and mRNA expressions of S1P1 and AT1R in normal liver tissues and human liver cancer tissues were determined with SP immuohistochemical method，Real-time PCR and Western blot.Results:Protein and mRNA expressions of S1P1 and AT1R in human liver cancer tissues was significantly stronger than that in normal liver tissues (P<0.05)，presenting a consistent trend.Conclusion:Protein and mRNA expressions of S1P1 and AT1R in human liver cancer tissues are enhanced，suggesting that there may be a correlation between the two molecules，and providing a new clue for further study of their relationship in the development of liver cancer，finally providing some new strategies for the prevention and treatment of liver cancer.
