miR-124在宫颈癌中的表达及对宫颈癌细胞增殖、迁移的影响

摘    要:目的:探讨miR-124在宫颈癌中的表达,及其对宫颈癌细胞增殖、迁移的影响。方法:收集宫颈癌组织和癌旁组织各13例,qRT-PCR法检测miR-124的表达,免疫组化检测STAT3的表达。生物信息学技术预测miR-124的靶基因,双荧光素酶报告基因、qRT-PCR、Western blot实验验证。每种宫颈细胞随机分为三组,一组转染miR-124 mimics,一组不转染,一组转染miR-124 inhibitor。CCK-8实验检测细胞增殖能力,细胞划痕实验检测细胞迁移能力。结果:miR-124在宫颈癌组织和细胞系中表达均降低(P均<0.05),STAT3在宫颈癌组织中的表达明显增高(P<0.05)。STAT3是miR-124的直接靶基因,miR-124可靶向调节STAT3的mRNA和蛋白表达水平(P均<0.05)。在Hela及Siha细胞中,miR-124 mimics转染组细胞增殖及迁移能力均降低(P均<0.05),miR-124 inhibitor转染组细胞增殖及迁移能力均增加(P均<0.05)。结论:miR-124在宫颈癌中低表达,可能通过靶向调节STAT3表达抑制宫颈癌细胞的增殖及迁移能力。

Abstract:Objective:To investigate the expression of miR-124 in cervical cancer and its effect on the proliferation and migration of cervical cancer cells.Methods:There were 13 cases of cervical cancer and adjacent tissues.The expression of miR-124 was detected by qRT-PCR.Immunohistochemistry was used to detect STAT3 expression.Bioinformatics techniques predicted the target genes of miR-124.Dual luciferase reporter gene,qRT-PCR,Western blot were used to validated.Each cervical cell was randomly divided into three groups.One group was transfected with miR-124 mimics.One group did not be transfected,and one group was transfected with miR-124 inhibitor.CCK-8 test was used to detect cell proliferation.Cell scratch assay detected cell migration ability.Results:miR-124 expression in cervical cancer tissues and cell lines were reduced(all P<0.05).STAT3 expression in cervical cancer tissue was significantly higher(P<0.05).STAT3 was a direct target of miR-124.miR-124 could target STAT3 mRNA and protein expression level(all P<0.05).The proliferation and migration ability of miR-124 mimics transfected cells in Hela and Siha cell lines were decreased(all P<0.05).miR-124 inhibitor transfected cells increased proliferation and migration ability(all P<0.05).Conclusion:miR-124 is low expressed in cervical cancer.It is possible to inhibit the proliferation and migration of cervical cancer cells by targeting STAT3 expression.
