响应面法优化香姜中6-姜辣素的提取工艺及其抗氧化活性研究

摘    要:目的:根据响应面法优化湖南永州江永香姜中6-姜辣素的提取工艺及其抗氧化活性研究。方法:对乙醇浓度、料液比、提取时间(t)进行单因素试验,运用Box-Behnken中心组合试验和响应面法考察乙醇浓度、料液比、提取时间对6-姜辣素得率的影响并以2,6-二叔丁基对甲酚和L-抗坏血酸作对照,测定香姜6-姜辣素总还原能力。结果:优化后的最佳提取工艺:乙醇体积分数为80%、料液比1:12(g·mL-1)、提取时间90 min,在此条件下,6-姜辣素得率达90%以上。香姜中6-姜辣素体外清除羟基自由基能力的IC50值为0.60 mg·mL-1结论:该法相对水提法、压榨法,该工艺更具提取率高,回收率高等优点。6-姜辣素总还原能力和清除羟自由基能力弱于同浓度的L-抗坏血酸(Vc),但强于2,6-二叔丁基对甲酚(BHT)。

Abstract:OBJECTIVE Response surface methodology was employed to optimize the extraction process of 6-gingerols collected from Jiangyong (Hunan province) and evaluate its antioxidant activity.METHODS The single factor tests of alcohol concentration, liquid-solid ratio and extraction time were firstly conducted. Then, the effects of alcohol concentration, liquid-solid ratio and extraction time (t) on extraction yield were studied by Box-Behnken test design and response surface method. Taking BHT and Vitamin C as the control to determine Xiangjiang 6-gingerol total reduction ability.RESULTS The optimal 6-gingerol extraction process:alcohol concentration 80%, liquid-solid ratio 1:12 (g·mL-1), extraction time (t) 90 min. Therefore, under these conditions, the extraction rate of 6-gingerol was up to 90%. The IC50 of the obtained 6-gingerol againsthydroxyl radicals was 0.60 mg·mL-1.CONCLUSION Relative water extraction and pressing method have the advantages of high extraction rate and high recovery rate. The reducing ability and scavenging impact of the radicals of 6-gingerol from ginger were weaker than those of Vitamin C at the same concentration, but still more effective than 2,6,-di-tert-butyl-4-methyl phenol (BHT).
