胃癌中EphB3的表达及其对癌细胞增殖、侵袭能力的影响

目的:研究EphB3在胃癌中的表达及其与临床病理特征的相关性，并探索EphB3对胃癌细胞增殖、侵袭能力的影响。方法:利用免疫组织化学染色法检测60例胃癌蜡块及配对的癌旁组织中EphB3的表达，并与临床病理特征进行相关性分析。转染过表达胃癌细胞株SGC7901中EphB3，通过CCK-8实验检测EphB3对胃癌细胞增殖能力的影响。采用细胞划痕实验观察EphB3对胃癌细胞侵袭能力的影响。结果:EphB3在胃癌中的阳性表达率明显低于癌旁组织（P=0.004），且与肿瘤分化程度、浸润深度、淋巴结转移存在相关性(P<0.05)。转入外源性的EphB3显著降低细胞的增殖、侵袭能力。结论:EphB3能够抑制胃癌发生、发展的过程，可能成为未来胃癌靶向治疗的重要研究方向。

The expression of EphB3 in gastric cancer and its influence on proliferation and invasion of cancer cells

Objective:To explore the expression of EphB3 in gastric cancer and its biological effects on cell proliferation and invasion.Methods:The expression of EphB3 in 60 specimens of gastric cancer tissues and 60 spceimens of adjacent tissues was detected by immunohistochemistry.The relationship between EphB3 expression and clinicopathological features was statistically analyzed.The biological effects of overexpression of EphB3 on proliferation，invasion of gastric cancer cell were detected by CCK-8 and wound-healing assay.Results:According to IHC analysis，positive rates of EphB3 protein were significantly lower in gastric cancer than in adjacent tissue (P=0.004).EphB3 expression was associated with tumor differentiation，invasion depth and lymph node metastasis (P<0.05).And EphB3 could inhibit proliferation and invasion in SGC7901 cell line.Conclusion:EphB3 can inhibit the tumorgenesis，progression of gastric cancer and may become an important research direction of targeted therapy for gastric cancer in the future.
