摘    要:目的:研究胆宁片中大黄素、大黄素甲醚和大黄酚对急性胆汁淤积大鼠的防治作用。方法:采用ɑ-异硫氰酸萘酯(ANIT)复制胆汁淤积模型,实验分为正常对照组、ANIT模型组(60 mg·kg-1)、胆宁片(3 g·kg-1)、大黄素(40 mg·kg-1)、大黄素甲醚(7 mg·kg-1)和大黄酚(4 mg·kg-1)保护组。除正常对照组和模型组其它各组大鼠均灌胃给予保护药,1次/天,连续9 d;第7次给保护药12 h后,模型组和其它各保护组动物均灌胃给予ANIT造模。造模48 h后观察各组大鼠胆汁流量变化,测定血清生化指标和检查肝脏病理组织形态。结果:胆宁片和大黄素能显著增加ANIT所抑制的大鼠胆汁流量;能显著降低ANIT导致的大鼠血清中谷丙转氨酶(ALT)、谷草转氨酶(AST)、γ-谷氨酰转氨酶(γ-GTP)、碱性磷酸酶(ALP)活性升高,以及逆转总胆红素(TBil)、直接胆红素(DBil)和总胆汁酸(TBA)的水平;而且胆宁片和大黄素能显著改善大鼠肝脏的组织形态。结论:胆宁片和大黄素对ANIT所致的大鼠急性胆汁淤积具有明显的防治作用,其中大黄素可能为胆宁片发挥利胆药效的有效成分之一。

Abstract:OBJECTIVE To assess the effects and possible action mechanisms of emodin, physcion and chrysophanol from Danning tablets on a-naphthylisothiocyanate (ANIT) induced liver injury with cholestasis.METHODS Danning tablets (3 g·kg-1·day-1), emodin (40 mg·kg-1·day-1), physcion (7 mg·kg-1·day-1) and chrysophanol (4 mg·kg-1·day-1) were intragastrically (i.g.) given to experimental rats for nine days. Rats were treated with ANIT (60 mg·kg-1 via i.g.) which caused liver injury with cholestasis at 12 h after the seventh administration of protective drugs. Bile flow, serum biochemical index and liver histopathological changes were measured at 48 h post ANIT treatment.RESULTS Obtained results demonstrated that Danning tablets and emodin significantly prevented ANIT induced suppression in bile flow, and reversed ANIT induced elevation of the serum levels of alanine aminotransferase (ALT), aspartate aminotransferase (AST), γ-glutamyltranspeptidase (γ-GTP), alkaline phosphatase (ALP), total bilirubin (TBil), direct bilirubin (DBil), total bile acid (TBA). Histopathology of the liver tissues showed that pathological injuries were relieved after Danning tablets and emodin treatment.CONCLUSION These findings indicate that Danning tablets and emodin exert preventive and protective effects against ANIT induced acute cholestasis in rats, and one of the possible protective components is likely emodin.
