三白草总黄酮对高脂血症大鼠血流变学及血管内皮功能的影响

摘    要:目的:探讨三白草总黄酮对高脂血症大鼠血流变学及血管内皮功能的影响。方法:雄性SD大鼠70只,随机分为正常组、模型组、阳性组(血脂康胶囊,100 mg·kg-1)和三白草总黄酮高(6.3 g·kg-1)、中(3.15 g·kg-1)、低剂量组(1.57 g·kg-1),正常组给予基础饲料喂养,其余各组喂养高脂饲料复制高脂血症大鼠模型,模型复制成功后开始药物干预治疗,治疗6周后测定各组大鼠血脂指标胆固醇(TC)、三酰甘油(TG)、低密度脂蛋白(LDL-C)、高密度脂蛋白(HDL-C)]、血流变学指标及血管内皮细胞因子的表达水平。结果:给药6周后,与模型组相比,三白草总黄酮各剂量组对大鼠TC、TG、LDL-C、TXB2、ET-1的表达水平有不同程度的降低,对HDL-C、6-keto-PGFla、NO表达水平有不同程度的升高,其中以三白草总黄酮的高剂量组作用效果最为明显,组间比较差异均有显著性(P<0.05);与模型组相比,三白草总黄酮各剂量组大鼠血浆黏度、血小板黏附率、全血黏度(高、中、低切)指标明显降低,组间比较差异显著性(P<0.05)。结论:三白草总黄酮能明显改善高脂血症模型大鼠的血脂水平,改善血流变学指标,使血液处于低浓、低黏、低聚、低凝状态,调节血管内皮功能,从而缓解血液流动阻力,增大血流量,促进机体的脂质代谢恢复正常。

Abstract:OBJECTIVE To observe the effects of total flavones from Saururus chinensis on hemorheology and vascular endothelial functions in hyperlipidemia rats.METHODS Seventy male Wister rats were randomly divided into six groups:the normal control group,model group,positive group (Xuezhikang capsule,100 mg·kg-1),high (6.3 g·kg-1),middle (3.15 g·kg-1),low dose (1.57 g·kg-1) total flavones groups.Total flavones were administered by gastric perfusion for 6 weeks.TC,TG,LDL-C,HDL-C,TXB2,6-keto-PGFla in serum of the rats were determined.Various indices of hemorheology were measured.RESULTS Compared with the model group,the total flavones from Saururus chinensis at different doses decreased the contents of TC,TG,LDL-C,TXB2,ET-1 in varying degrees,and increased the contents of HDL-C,6-keto-PGFla,NO in varying degrees at the end of the gastric perfusion.The effects of high dose total flavones were the most significant (P<0.05).Total flavones also had obvious effects on decreasing plasma viscosity,low,medium and high shear whole blood viscosity (P<0.05).CONCLUSION The total flavones from Saururus chinensis can significantly improve the blood lipid level of hyperlipidemia model rats,improve the indexes of hemorheology,result in low concentration,low viscosity,low coagulation,regulate endothelial function,thus relive blood flow resistance,increase blood flow,and promote the body's lipid metabolism to return to normal.
