冰片配伍红花对大鼠大脑中动脉脑缺血的影响及其机制研究

摘    要:目的:探讨冰片配伍红花对大脑中动脉脑缺血(middle cerebral artery occlusion,MCAO)大鼠的作用及其可能的机制。方法:雄性SD大鼠随机分为:假手术组、模型组、冰片组(0.03g·kg-1)、红花组(5g·kg-1)、冰片配伍红花组(冰片0.03g·kg-1+红花5g·kg-1)、尼莫地平组(0.01g·kg-1)。制备MCAO大鼠模型,术后连续灌胃给药7d对各组大鼠进行神经功能评分,激光多普勒仪检测大脑皮层脑血流量,TTC染色检测脑梗死范围,HE染色观察脑组织病理形态,免疫组化法检测脑组织MMP-2、MMP-9、ZO-1的表达,RT-PCR法检测MMP-9和Claudin-5的mRNA表达。结果:与模型组比较,冰片配伍红花组能降低大鼠神经功能评分,增加大脑皮层脑血流量,缩小脑梗塞范围,改善脑组织的病理形态,降低脑组织中MMP-2和MMP-9的表达同时提高ZO-1、Clauidin-5的表达。结论:冰片配伍红花对脑缺血大鼠具有脑保护作用,其脑保护的作用机制可能与下调MMP-2、MMP-9的表达和提高ZO-1、Claudin-5的表达有关。

Abstract:OBJECTIVE To investigate the effects of borneol combined with safflower on middle cerebral artery occlusion (MCAO) in rats and its possible mechanisms.METHODS The male SD rats were randomly divided into the sham operation group, the model group, the borneol group (0.03 g·kg-1), the safflower group (5 g·kg-1), the borneol combined with safflower group (0.03 g·kg-1+5 g·kg-1) and the nimodipine group (0.01 g·kg-1). The cerebral ischemia model rat was established by MCAO, the model rats were given different drugs through intragastric administration for 7 d, the neurological function scores were observed by Zea Longa method, cerebral cortical cerebral blood flow was detected by laser Doppler flowmetry, cerebral infarction were detected by TTC staining, histopathological changes of brain tissue were observed by HE staining, expressions of MMP-2, MMP-9 and ZO-1 were determined by immunohistochemical staining, mRNA expression of MMP-9 and claudin-5 were detected by RT-PCR.RESULTS Compared with the model group, the borneol combined with safflower group could significantly reduced neurological score, increased cerebral cortical cerebral blood flow, reduced the range of cerebral infarction, and improved the pathological form of brain tissue. The expressions of MMP-2 and MMP-9 in brain tissues were decreased, and expressions of ZO-1 and Claudin-5 were increased.CONCLUSION The borneol combined with safflower has protective effects on cerebral ischemia injury in rats, which is probably related to down-regulated expression of MMP-2 and MMP-9 and increased expression of ZO-1 and Claudin-5.
