摘    要:目的:探讨小剂量人参多糖(ginseng polysaccharides,GPS)对人肝癌BEL-7402细胞分化的诱导作用。方法:体外培养人肝癌BEL-7402细胞,GPS组加入10 mg·L-1人参多糖孵育,同时设对照组,采用CCK8实验检测人肝癌BEL-7402细胞增殖情况;显微镜、透射电镜观察细胞形态及亚细胞结构的变化;放射免疫法检测甲胎蛋白(AFP)、白蛋白(ALB)的分泌量;流式细胞术检测细胞周期。结果:人参多糖能抑制人肝癌BEL-7402细胞的增殖(P<0.05);人参多糖处理人肝癌BEL-7402细胞后细胞形态及亚细胞结构趋于成熟分化;甲胎蛋白(AFP)分泌量明显下降,白蛋白(ALB)的分泌量明显升高,与对照组比较差异具有统计学意义(P<0.05);GPS组G1期细胞比例显著高于对照组(P<0.05),细胞被阻滞G0/G1期。结论:小剂量人参多糖对人肝癌BEL-7402细胞生长有抑制作用,可能与诱导分化有关。

Abstract:OBJECTIVE To perform a series of experiments to explore the induction of cell differentiation of BEL-7402 cells by low dose ginseng polysaccharides (GPS).METHODS Human hepatoma BEL-7402 cells were cultured in vitro. The BEL-7402 cells were incubated with 10 mg·L-1 GPS. And the untreated BEL-7402 cells were used as controls. The cell proliferation of BEL-7402 was detected by CCK8. The cell morphologies and subcellular structures were characterized by scanning electron microscopy (SEM).The secretion of alpha-fetoprotein (AFP) and albumin (ALB) were determination by radioimmunoassay. Cell cycle was detected by flow cytometry.RESULTS GPS effectively inhibited the proliferation of BEL-7402 within certain concentration range (P<0.05), and the BEL-7402 cells incubated with GPS tended to mature and differentiate. The secretion of AFP was significantly decreased, while the secretion of ALB was significantly increased. Contrasted with the control groups, the difference was highly and statistically significant. G1 phase cell ratio of GPS group was significantly higher than that of control groups (P<0.05).CONCLUSION Low dose ginseng polysaccharides inhibit cell growth of BEL-7402 cells, which may be related to differentiation.
