| 摘 要： | 目的:探讨Bcl-2基因的RNA干扰(RNA interference,RNAi)表达载体对卵巢癌SKOV3细胞Bcl-2基因表达的抑制作用。方法:构建针对Bcl-2基因的RNAi表达载体,脂质体法转染卵巢癌SKOV3细胞株,应用RT-PCR及流式细胞仪检测其对Bcl-2基因mRNA及蛋白表达的影响。结果:构建的两种表达载体均抑制了Bcl-2基因的mRNA及蛋白的表达,其中转染2号载体的SKOV3细胞Bcl-2基因mRNA表达仅相当于对照组的32.3%,蛋白抑制率达60.18%。结论:RNAi表达载体可以有效抑制Bcl-2基因的表达。 |

| Abstract: | Objective:To investigate the inhibitory effects of RNAi expression vector of Bcl-2 gene on Bcl-2 gene expression of SKOV3 cell of ovarian carcinoma.Methods:RNAi expression vector for Bcl-2 gene was constructed and transfected to SKOV3 cell strain of ovarian carcinoma with liposomes. RT-PCR and flow cytometer detects effect to mRNA and protein expression of Bcl-2.Results:The two kinds of constructive expression vectors inhibited both mRNA and protein expression of Bcl-2. Among Bcl-2 mRNA and protein expression of SKOV3 cell that transfected B type carrier only equaled to 32.3%of contrastive groups. The anti-rate of protein was 60.18%.Conclusion:The RNAi expression vector inhibits effectually expression of Bcl-2 gene. |
