高温与治癌药物联合应用对人肺腺癌细胞的杀伤作用

摘    要:目的 观察高温和丝裂霉素(MMC)、长春酰胺(VDS)及顺铂(DDP)联合应用对人肺腺癌细胞的杀伤作用。方法 将实验对象分为对照组、高温组、药物组、药物联合高温组。采用细胞生长曲线及集落形成试验检测细胞增殖动态变化、流式细胞术检测细胞周期及凋亡、电镜观察细胞形态、医学多功能图像分析系统检测细胞核仁组织区银染(Ag-NOR3)含量。结果 细胞增殖动态变化显示,药物和/或高温处理细胞生长明显受抑,其中以药物联合高温作用最为明显;流式细胞术分析细胞周期显示,药物联合高温组在24h出现S期阻滞,S期细胞达88.18%,并明显出现细胞凋亡小峰;细胞形态学变化显示,药物或联合高温组24h引起明显的细胞损伤,细胞空泡多,特别是药物联合高温组能进一步使细胞核碎裂,出现典型的细胞凋亡变化;细胞核内Ag-NOR3含量变化结果显示,药物联合高温处理后24h,Ag-NOR3在镜下观察,均显示核内黑色颗粒,胞核呈淡黄色,对比鲜明。结论 高温与抗癌药物对人肺腺癌细胞有协同杀伤作用。

Abstract:Objective To observe the kill effect of hyperthermia combined with anti-cancer drugs(mitomycin,vindesine,cisplatin) on human lung adenocarcinoma cell lines(D6).Methods Experimental objects were divided into four groups:Control group,hyerthermia group,chemotherapy group and chemohyperthermia group.Dynamic change of cell proliferation was detect by cell growth curve and clone formation assay;Cell cycle and apoptosis were test by flow cytometry;Cell morphology was observed by electron microscope;Ag-NOr3 in nucleus was determined by multi-function image analysis system.Results After treatment,the dynamic change of cell proliferation all was obviously inhibited in chemotherapy group,hyperthermia group,particularly in the chemohyperthermia group.Of cell cycle,the blockage in S-phase was found at 24 hours after chemohyperthermia group,it reached 88.18%,small apoptotic peak was also presented.There found obvious cell damage and cell vacuolation after 24 hours of chemotherapy or chemohyperthermia treatment.Even as cell disruption to typic apoptosis was appeared in chemohyperthermia group.While the Ag-NOR3 in nucleus appeared black granules,nuclei got yellow under the electron microscope.Conclusion The study suggests that the hyperthermia combined with anti-cancer drugs has a kill effect for treating the human lung adenocarcinoma cells.
