钛金属材料表面微形态对兔成骨细胞生物学行为的影响

摘    要:目的 体外观察钛金属材料不同表面微形态对兔成骨细胞生物学行为的影响.方法 采用激光扫描、喷砂、机械加工和抛光技术在钛金属基片上制备四种不同的表面微形态,激光扫描表面(LS)为50μm宽度的平行微沟槽,机械加工表面(MS)为100 μm宽度大环形沟槽,两种表面均为规则纹理,喷砂表面(SS)为完全粗糙表面,抛光表面(PS)为光滑表面;扫描电镜观察表面形态特征并测定表面形态的粗糙度(Ra),分别在不同基片表面接种兔成骨细胞,培养12 d,测定细胞24 h黏附情况、12 d增殖情况、11 d碱性磷酸酶活性,并通过扫描电镜和荧光显微镜观察细胞形态,统计分析各组间的差异.结果 细胞黏附LS=SS>MS=PS,细胞增殖SS>LS>MS=PS,ALP活性LS=SS>MS=PS,在LS和SS表面上的细胞黏附、增殖及ALP活性均高于MS和PS表面(P<0.05).结论 钛金属材料表面微形态对兔成骨细胞的黏附、增殖、分化以及细胞形态产生影响,细胞级别的规则微沟槽表面为成骨细胞提供了一个更适合的生长环境.

Effect of surface micro-topography of titanium material on biological behavior of rabbit osteoblasts in vitro
Abstract:Objective To investigate effects of surface micro-topography of titanium material on biological behavior of rabbit osteoblasts in vitro.Methods Four different types of surface mi- cro-topography were created on titanium material by laser-scanning,sandblasting,machine-tooling and pol- ishing techniques.The laser-scanned surface (LS)and the machine-tooled surface (MS)showed regular suifaee texture.Widths of their grooves were 50 μm and 100 μm respectively.The sandblasted surface(SS) was rough and the polished surface (PS) was smooth.Scanning electron microscopy was used to observe surface topographical features and to measure the roughness amplitude(Ra).The titanium samples were seeded with rabbit osteoblasts and maintained for 12 days.Adhesion at 24 h,proliferation at 12 days and ALP activity at 11 days were assessed.The cell morphology was observed by scanning electron microscopy and fluo- rescence microscopy.Results Cell adhesion Was LS=SS> MS=PS,cell proliferation was SS>LS> MS=PS and ALP activity Wag LS=SS>MS=Ps.The cell adhesion.proliferation and ALP activity on LS and SS were higher than on MS and PS(P<0.05). Conclusions Surface micro-topography of ti- tanium material can influence adhesion.proliferation.differentiation and morphology of rabbit osteoblasts in vitro.Regular micro-grooved titanium surface at the cell scale can provide a better cell growth enviromnent.
