锁阳对大鼠肝纤维化干预作用的实验研究

摘    要:目的:观察锁阳对四氯化碳(CCl4)诱导的大鼠肝纤维化模型的预防及逆转作用。方法将SD大鼠按照随机数字表法分为正常对照组、模型组、锁阳干预Ⅰ组、锁阳干预Ⅱ组和锁阳对照组5组,每组各10只。CCl4与花生油混合液造模,根据分组在不同时间给予锁阳灌胃。实验结束后,取血清和肝组织,检测血清肝功能,HE染色和M asson染色,观察肝组织病理改变,流式细胞仪测大鼠血浆 CD3+、CD4+和CD8+水平。结果模型组大鼠肝组织纤维化分级主要以++++级为主,锁阳干预Ⅰ组分级主要以++、+++级为主,锁阳干预Ⅱ组分级主要以+、++为主。与正常对照组比较,模型组大鼠血清AST、ALT活性明显升高,CD3+、CD4+水平降低,CD8+水平升高,肝脏指数升高。与模型组相比,锁阳干预Ⅰ组大鼠血清中ALT活性降低,CD3+、CD4+水平升高,CD8+水平降低;锁阳干预Ⅱ组中大鼠血清中AST、ALT活性明显降低,CD3+水平升高,CD8+水平降低;两组大鼠肝脏指数均降低,差异均有统计学意义(P<0.05)。结论锁阳对肝纤维化有一定的预防和逆转作用,同时能影响肝纤维化大鼠外周血T淋巴细胞亚群,在一定程度上拮抗免疫功能低下。

Abstract:Objective To observe the preventive and reversal effects of Cynomorium songaricum on liver fibrosis of rats induced by carbon tetrachloride (CCl4 ) .Methods SD rats were randomly divided into the normal control group ,the model group ,the Cynomorium songaricum intervention Ⅰ group ,the Cynomorium songaricum intervention Ⅱ group and the Cynomorium songaricum control group .Each group consisted of 10 rats ,which were injected subcutaneously with the mixture of carbon tetrachloride and peanut oil to make the models of liver fibrosis . Then the rats were given Cynomorium songaricum via intragastric administration at different time in the different groups respectively .Serum and livers were collected after all the rats were sacrificed at the end of the experiment . Hematoxylin and eosin red (HE) staining and Masson staining were used to observe the pathological changes of liver tissue ,and flow cytometry was used to measure the levels of CD3+ ,CD4+ and CD8+ in the plasma .Results According to the grading of liver fibrosis ,liver tissue of the rats in the model group mainly belong to grade ++++ , the liver tissue of the rats in the Cynomorium songaricum intervention Ⅰ group were mainly in grade ++ and grade+++ and the liver tissue of the rats in the Cynomorium songaricum intervention Ⅱ group were mainly in grade +and grade ++ .Compared with the normal control group ,the activities of AST and ALT in the serum of the rats in the model group were significantly increased ,the levels of CD3+ and CD4+ were reduced ,the level of CD8+ was increased ,and the liver index was elevated .Compared with the model group ,the activities of ALT in the serum of rats in the Cynomorium songaricum intervention Ⅰ group was lower ,the levels of CD3+ and CD4+ were higher ,and the level of CD8+ was lower ;The activities of AST and ALT in the serum of rats in Cynomorium songaricum intervention Ⅱ group was lower ,the levels of CD3+ and CD4+ were higher ,and the level of CD8+ was lower ;The liver index of rats in both intervention groups were decreased .All the differences mentioned above were statistically significant (P< 0.05) .Conclusion Cynomorium songaricum has good preventive and reversal effects on liver fibrosis .Meanwhile ,it has certain effects on T lymphocyte subgroups in peripheral blood of rats with liver fibrosis and it can improve their immune hypofunction to a certain degree .
