摘    要:目的建立监测人全血中环孢素A质量浓度的反相高效液相色谱(RP-HPLC)法。方法采用反相C18色谱柱,流动相为乙腈-甲醇-水(160:30-15),柱温70℃,流速0、8mL/min,检测波长210nm。结果环孢素A质量浓度在15—1600ng/mL范围内与内标的峰高比有良好的线性关系(r=0.9993),最低检测限为5ng/mL,高、中、低3种质量浓度平均回收率为96.02%~98.31%,日内精密度RSD为1.24%~3.37%(n=5),日间精密度RSD为3.45%~4.27%(n=5)。结论该法简单、准确、经济,可用于环孢素A的血药浓度监测。

Abstract:Objective To establish a method to detect the concentration of cyclosporine A in human whole blood, Methods C18 column was adopted and the temperature was set at 70℃.The mobile phase was a mixture of acetonitrile, methanol and water (160:30:15) and the flow rate was 0. 8mL/min. The UV detector was set at 210nm. Results The calibration curve was linear in the concentration range from 15 ng/mL to 1 600 ng/mL in blood (r=0, 999 3). Within the range,the recovery rate of low,moderate and high concentration was 96.02%-98.31%.The lowest limit of quantitation was 5 ng/mL. The intraday precision was 1.24%-3.37% (n=5).The interday precision was 3.45% -4. 27% (n = 5). Conclusion This method is easy, convenient and precise to detect the concentration of cyclosporin A in blood.
