挤压伤后脊髓腹角运动神经元NGF,BDNF表达的变化

摘    要:为探讨脊髓挤压伤后早期不同时间腹角运动神经元NGF和BDNF的表达变化,建立脊髓挤压伤模型,取挤压伤位点尾侧段T13节段脊髓制作冰冻切片,运用NGF,BDNF兔抗血清以免疫组化ABC法染片,观察并计数腹角NGF和BNDF的阳性神经元数。结果:NGF主要分布于正常大鼠脊髓腹角运动神经元的胞核及胞浆,损伤后21d,NGF的阳性神经元数较对照、损伤1d及7d组均明显增多(P<0.01),与NGF比较,对照组脊髓腹角亦见BDNF免疫阳性神经元,胞浆染色、胞核不着色,损伤后24h,BDNF阳性神经元数已较正常者有显著增加(P<0.05),此后维持该水平至21d,结论:脊髓挤压伤后腹角神经元NGF和BDNF的表达明显增加,提示NGF和BDNF可能在脊髓损伤修复的早期反应中发挥作用。

Abstract:Objectve: to explore the changes of NGF and BDNF in neurons of ventral horn in different time following crushed spinal cord injury(cSCI). Method: immunohistochemical ABC method was used to explore the changes of NGF and BDNF in neurons of ventral horn in different time following cSCI. Results: the results showed that NGF and BDNF positive products were mainly observed in neurons of ventral horn in intact group. After cSCI, Although the number of NGF positive neurons increased in 1?d group and 7d group, there is no statistical significance compared with intact group(P>0 05). However, in 21?d group following injury, the numbers of NGF positive neurons more apparently increased than those of control group, 24?h group and 7?d group(P<0 05). Compared with NGF, the numbers of BDNF positive neurons more greatly increased in 24?h following injury than those of control group and this change was kept to 21?d following injury. Conclusion: the expression of NGF and BDNF was upregulated at different time following injury. It indicated that NGF and BDNF not only involves in the physiological function of spinal cord in adult rat but also may play an important role in the processes of nerve injury following cSCI.
