摘    要:目的:明确左归丸中总黄酮的提取方法及其含量,为复方左归丸的进一步研究、开发利用提供科学依据。方法:用均匀设计U5(53)表安排左归丸总黄酮的多因素多水平提取实验,以浸膏得率为指标,确定醇提左归丸的数学模型,得出左归丸醇提浸膏得率与各因素之间的回归关系,并推算最佳提取工艺条件。并应用AlCl3-KAc法显色,利用分光光度法以芦丁为对照品对浸膏中总黄酮进行含量测定,对均匀设计结果进行验证。结果:醇提左归丸的最佳工艺条件为:复方左归丸混合粉末质量(g)与加入乙醇体积(mL)的配比=1∶12,乙醇体积分数为55%,提取时间为80 min,浸膏得率的最大值为(69.8±9.62)%。65%乙醇提取左归丸所得浸膏中总黄酮含量最高。结论:用U5(53)法得到的模型方程可在设立的因素水平范围内给出醇提左归丸浸膏得率的极大值,进而探讨各因素对醇提左归丸浸膏得率的影响机制。

Abstract:Objective:To provide evidence for the future exploration and development of the Zuoguiwan though clear the extraction approach and the content of the total flavones component of the Zuoguiwan. Method: The experiment for extract- ing the total flavones of zuoguiwan with factors and multiple levels was designed by uniform design according to U5(53) and its usage table. The mathematic model for the extraction of the total flavones of zuoguiwan was required to show the correlation between the total flavones of zuoguiwan and three factors, and the optimal technological conditions were ob- tained. Result:The optimum extraction technology is:the ratio of solid to liquid being 1:12, and extracting time being 80 minutes.Under the optimal condition,the yield rate of the total flavones is predicted to be(69.8±9.62)%. Conclusion:By using U5(5^3), the optimal yield rate of the total flavones of zuoguiwan can be calculated by the mathematic model within experiment range, and then the factors affecting the extraction of the total flavones of zuoguiwan can also be investigated.
