尿中2,5-己二酮生物限值的探讨及方法学研究

摘    要:目的推荐我国非职业接触正己烷者尿中2,5-己二酮生物限值并建立相应的气相色谱测定方法。方法选择300名无职业性正己烷接触史的健康成人,其中吸烟者80人,不吸烟者220人。将尿样经盐酸水解,用二氯甲烷萃取、FFAP色谱柱分离,氢火焰离子化检测器检测,以保留时间定性,峰面积定量。结果对深圳市300名健康成人尿中2,5-己二酮进行了测定,男女2组经t检验,差异无统计学意义(t=0.838,P〉0.05)。吸烟组与不吸烟组者经t检验,差异无统计学意义(t=1.40,P〉0.05)。我们用气相色谱法测得的结果其测定范围0.00-60.62μzmol/L,平均浓度为23.74μmol/L,相对标准偏差为1.554,95%上限为57.21μmol/L。结论建议深圳市健康成人尿2,5-己二酮生物限值上限为57.21μmol/L。

Abstract: Objective ] To recommend the national biological limit value of 2,5-hexanedione in urine of non-occupational exposure, and establish appropriate method of gas chromatography. Methods] 300 healthy adults, who had no occupational exposure history were selected, including 80 smokers and 220 non-smokerso The urine samples were extracted by diehloromethane after hydrolyzed by hydrochloric acid, then were separated by FFAP chromatographic column. After that the samples were tested by flame ionization de- tector (FID) to retain the time character and qualified by peak area. Results] 300 samples of 2, 5-hexanedione in healthy adults urine were detected. T test was applied to analysis the data between male and female groups, the difference was no significant ( t = 0. 838 ,P 〉0.05). And there was no significant difference between the smoker group and non-smoker group neither ( t = 1.40, P 〉 0.05 ). The testing results range of gas chromatography was 0.00 - 60.62 μ mol/L. The average concentration was 23.74 μ mol / L, standard deviation was 1. 554, the high limit was 57.21 p, mol/L. Conclusion] It is recommend that the high limit value of 2 ,5-hexanedione in urine is 57.21 μ mol/L in healthy adults in Shenzhen city.
