摘    要:目的:探讨人表皮生长因子受体(EGFR)、Kirsten鼠肉瘤基因(K-ras)和核苷酸剪切修复复合体1(ERCC1)蛋白在非小细胞肺癌(NSCLC)组织和癌旁组织中的表达情况及其与NSCLC生理病理特征的相关性。方珐:用免疫组化方法检测NSCLC患者手术切除标本89例,其中取32例患者癌旁组织标本,检测各标本中EGFR、K—Ras和ERCC1蛋白的表达。结果:NSCLC组织中EGFR蛋白阳性表达率为52.8%,在癌旁组织中阳性表达率为3.12%;Ⅰ-Ⅱ期、无淋巴结转移患者中阳性表达率明显低于Ⅲ-Ⅳ期、有淋巴结转移患者;K—Ras蛋白阳性表达率为41.5%,在癌旁组织中为9.4%;腺癌表达率明显高于鳞癌;ERCC1蛋白阳性表达率为44.9%,在癌旁组织中为3.12%,差异均具有统计学意义(P〈0.05)。结论:NSCLC患者中EGFR、K—Ras和ERCC1蛋白的表达均为上调趋势,其表达是指导患者临床个体化疗方案及疗效和预后评价的重要指标。

Abstract:Objective: To detect expression of EGFR, K-Ras and ERCC1 protein in tissues of non-small cell lung cancer (NSCLC) and adjacent tissues and to analyze the correlation between the expression level and of pathologic parameters of the disease. Methods: expression of EGFR, K-Ras and ERCC1 protein in eighty-nine specimens of resected tissues of confirmed lung cancer and twenty-nine specimens of adjacent tissues of these lung cases were detected by immunohistochemical method. Results: Positive expression rate of EGFR protein in NSCLC tissues and adjacent tissues were 52.8% and 3.12%, respectively, EGFR expression rate in stage Ⅰ -Ⅱ NSCLC without lymph node metastasis were significant lower than that in stage Ⅲ-Ⅳ cases with lymph node metastasis. Positive expression rate of K-Ras protein was 41.5% in NSCLC tissues and was 9.4% in the adjacent tissues, and the expression rate was higher in adenocarcinoma than that in squamous cell carcinoma. Positive expression rate of ERCC1 protein was 44.9% in NSCLC tissues and was 3.12% in the adjacent tissues, the difference was significant (P〈0. 05). Conclusion: The expression of EGFR,K-Ras and ERCC1 protein was up-regulated in NSCLC indicating they are important indicators for individual chemotherapy and evaluation of prognosis.
