| Abstract: | Objective: To establish a rapid, sensitive and specific dot immunogold filtration assay (DIGFA) for the detection of hepatitis A virus specific immunoglobulin M (IgM) antibody. Methods: Hepatitis A antigen, which was used to capture the anti HAV IgM from sera, was dotted on nitrocellulose membrane. The IgM was revealed by using anti human IgM antibodies conjugated to colloidal gold particles. The procedure could be finished in 2 to 4 min. The color of the positive dots was bright enough to be determined. Results: 279 serum samples were comparatively detected with both DIGFA and ELISA. 148 of the 279 serum samples were positive and 125 were negative with both methods. The coincidental rate was 97.8%.The specificity of DIGFA was confirmed by blocking test and 2 ME treatment. The DIGEA does not require sophisticated equipment and is not interfered with rheumatoid factor in sera. Conclusion: The method could be applied for early diagnosis and epidemiological survey of hepatitis A. |
