IGF1和IGF2表达与非小细胞肺癌淋巴结转移的相关性

摘    要:目的研究胰岛素样生长因子1和2(IGF1、IGF2,合称为IGFs)在非小细胞肺癌(NSCLC)组织中的表达,探讨其与NSCLC区域淋巴结转移的关系。方法采用免疫组织化学和免疫印迹技术检测IGF1和IGF2在65例NSCLC、29例癌旁肺组织和19例良性肺病变组织中的表达。结果IGF1和IGF2在NSCLC中的表达率显著高于良性肺组织。IGF1和IGF2在伴有区域淋巴结转移肺癌中的表达显著高于无淋巴结转移肺癌组织。IGF1和IGF2在伴有N2或≥5个转移淋巴结肺癌组织中的表达显著高于在仅伴有N1或<5个转移淋巴结肺癌组织。IGF2在伴有多组转移淋巴结肺癌组织中的表达显著高于仅有一组转移淋巴结肺癌组织。结论IGFs不仅在NSCLC中过表达,而且与区域淋巴结转移的部位、数量及组数相关联,提示IGFs在NSCLC发生和发展中可能起重要作用,IGFs为NSCLC生物学行为尤其是淋巴结转移的判断提供一个新的有意义的指标。

Abstract:Objective To investigate the expression of insulin-like growth factors (IGF1 and IGF2, or IGFs) in non-small cell lung cancer (NSCLC) as well as the correlation between IGFs expression and regional lymph node metastasis. Methods The expression of IGF1 and IGF2 was investigated in 65 specimens from patients with NSCLC (29 cases contained normal lung tissue adjacent to cancer) and in 19 specimens from patients with benign lung diseases by immunohistochemistry and western blotting. Results IGF1 and IGF2 were overexpressed in NSCLC compared to benign lung tissues. The expression of IGF1 and IGF2 was significantly higher in NSCLC with metastasis to regional lymph node than those without metastasis to regional lymph node. Furthermore, the expression of IGF1 and IGF2 was significantly higher in NSCLC with N2-stage or with metastasis to 5 or more regional lymph nodes than those only with N1-stage or with metastasis to less than 5 regional lymph nodes. In addition, IGF2 expression was statistically higher in NSCLC with multiple groups of metastatic regional lymph node than those only with single group of metastatic regional lymph node. Conclusion The overexpression of IGFs in NSCLC was positively correlative to lymph node metastasis, with respect to the stations, numbers and groups of metastatic regional lymph node, it indicates that IGFs may play an important role in the initiation and progression of NSCLC; the expression of IGFs may be valuable in evaluation of the biological behavior of NSCLC.
