摘 要: | 目的 探讨喉癌患者外周血CD3T细胞分泌细胞因子的水平变化,分析Th1和Th2细胞的细胞因子免疫活动。为肿瘤的免疫治疗提供实验依据。方法采用刺激物刺激细胞,增加细胞内细胞因子的表达,再加入荧光标记的特异性抗细胞因子单克隆抗体,特异性抗原抗体结合,最后应用流式细胞仪分析特异性细胞因子表达水平。用酶联免疫吸附法(ELISA)检测相应的细胞因子的方法进行研究。结果喉癌患者Th1型细胞因子干扰素-γ(IFN-γ)、白细胞介素-2(IL-2)、白细胞介素-12(IL-12)表达水平较正常对照组显著降低,Th2型细胞因子白细胞介素-4(IL-4)和白细胞介素-10(IL-10)表达水平较正常对照组升高,差异有统计学意义。肿瘤坏死因子-α(TNF—α)表达水平较正常对照组升高,差异有显著性。喉癌患者体内Th2型细胞因子模式占优势状态。结论肿瘤细胞发生免疫逃逸,从而导致肿瘤的发生或者转移的原因之一。

Abstract: | Objective To detects the level of Th1 and Th2 type cytokines of peripheral blood CD3+ T lymphoeytes in the patients with laryngeal neoplasmy. Methods Peripheral blood lymphoeytes were stimulated to enhance expression of cvtokines. With specific fluorochrome-conjugated monoclonal antibody combined with antigen. Cytokines were detected by flow cytometry and ELISA. Results The levd of Th1 type cytokines such as interferon-γ (IFN-γ),interleukin2 (IL-2) interleukin12 (IL-12) were significantly lower in patients of laryngeal neoplasm than those of normal controls. The levels of Th2 type cytokines such as interleukin-4(IL-4),interleukin-10(IL=10) were higher in patients of laryngeal neoplasm than those of normal controls,showing significant difference .The levels of tumor necrosis factor a (TNF-α)in patients were significant higher than normal control. Conclusion The levels of Th1/Th2 were varied in laryngeal neoplasm Thl type cytokines were inhibited,and Th2 type cytokines were relatively enhanced.The mechanism of tumor occurrence and metastasis may be caused by immune escape from immune surveillance. |
