胃癌中Cks1 、p27 Kip1和Skp2 蛋白的表达

 目的 探讨Cks1在胃癌发生及其在Skp2调节p27 ^Kip1降解过程中的作用。方法 应用流式细胞术测定正常胃粘膜和胃癌组织中Cks1、p27^Kip1、Skp2蛋白表达。结果胃癌组织中Cks1、Skp2表达明显高于正常胃粘膜（x²=22．69，P〈0．05；x²=13．42，P〈0．05），而p27 ^Kip1表达则低于正常胃粘膜（x²=14．83，P〈0．05）。胃癌中Cks1、Skp2表达与p27 ^Kip1表达呈负相关（r=-0．649，P〈0．05；r=-0．732，P〈0．05）；而Cks1蛋白表达与Skp2蛋白缺乏相关性（P〉0．05）。胃癌中Cks1的表达与肿瘤分化程度相关（x²=5．05，P〈0．05），而与肿瘤浸润深度、淋巴结转移及临床分期不相关（P〉0．05）。结论Cks1可能参与了胃癌的发生；胃癌中Cks1可能参与了Skp2调节p27 ^Kip1泛素化降解过程。

Correlation of Cks1, Skp2 and p27Kip1 Protein Expression in Gastric Cancer

Objective 　To investigate the expression of Cks1 protein and its relationship with the expression of Skp2 , p27 ^Kip1 and tumor aggressiveness. Methods 　Flow cytomet ry was used to detect the expression of the three proteins in 20 cases of normal gast ric tissue and 64 cases of primary gastric cancers. Results 　The expression of Cks1 and Skp2 in gastric cancers was higher than that in normal gast ric tissue (χ² = 22. 69 , Ρ< 0. 05 ;χ² = 13. 42 , Ρ< 0. 05 respectively) . While the expression of p27 ^Kip1 was lower than that in normal gastric tissue (χ² = 14. 83 , P < 0. 05) . A significant inverse relation was observed between the expression of Cks1 or Skp2 and of p27 ^Kip1 ( r = - 0. 649 , P < 0. 05 ; r = - 0. 732 , P < 0. 05 , respectively) . While there was no correlation between the expression of Cks1 and Skp2 ( P > 0. 05) . The expression of Cks1 was significantly associated with tumor differentiation. (χ2 = 5. 05 , Ρ< 0. 05) . While it was not associated with depth of invasion , lymph node metasis and clinical stages ( P > 0. 05) . Conclusion Cks1 may play an important role in human gast ric tumorgenesis and it may be an essential accessory factor for Skp2 in protease-dependent dest ruction of p27 ^Kip1.
