抗肿瘤和乙肝双交IL18—HBV S基因疫苗的构建

目的：探讨IL－18及乙肝病毒S基因（HBVS）的联合功效。方法：从胎肝组织中抽提总RNA，RT－PCR扩增IL－18cDNA基因。从载体pcDNA3．0／S中限制性酶切获取HBV S基因，然后，将其与IL－18cDNA一半亚克隆到真核表达质粒pIRES-EGFP中。结果：构建了具有抗肿瘤及乙肝双重功效的IL－18－HBV S乙肝基因疫苗。结论：含有IL－18基因的HBV基因疫苗的构建为基因疫苗的功能和应用研究提供了基础。

Recombination of IL18-HBV S Gene Vaccine to Resist Tumor and Hepatitis B

To find the combination function of IL_18 and HBV S gene. IL_18 cDNA was obtained by RT_PCR from human embryo liver tissue. And an eukaryote expression vector _pEGFP_N1/IL_18 with a report gene which was expressing green fluorescene protein was constructed. Then the authors got the IL_18 cDNA by restriction enzyme and PCR.Because of its liverphagy and immunization ,the authors used HBV S gene from an expression vector_pcDNA3.0/S and linked it with IL_18 cDNA . This two genes were subcloned into an expression eukaryote vector_pIRES_EGFP and formed a new expression vector pIRES_EGFP_S_IL18 that would resist tumor and Hepatitis B.[Conclusion]The recombination of IL18_HBV S gene makes great progress in enhancing the immunity of HBV gene vaccine.It could play a basic role in resisted tumor and Hepatitis B.
