指状青霉致突变性研究

目的：研究指状青霉（Ｐｅｎｉｃｉｌｌｉｕｍ ｄｉｇｉｔａｔｕｍ）提取物的致突变性。方法：采用了细菌回复突变试验，小鼠骨髓 ＰＣＥ微核试验，大鼠肺及肝原代细胞程序外ＤＮＡ合成试验（ＵＤＳ）和大肠杆菌 Ｋ１２ ｉｎｆａ基因突变试验及 ｉｎｆＡ基因 序列测定和分析。结果：指状青霉提取物：①可明显地诱发大肠杆菌ＮＤ－１６０菌株回复突变；②可使小鼠骨髓 ＰＣＥ微核率极显著增高（Ｐ＜０．０１）；③可显著诱导大鼠肝原代细胞ＵＤＳ（Ｐ＜０．０５）及极显著诱导肺原代细胞ＵＤＳ （Ｐ＜０．０１）；④可诱发大肠杆菌Ｋ１２ ｉｎｆＡ基因ＤＮＡ序列中５个碱基位点突变，且其中１个位点的突变还可导致翻 译相应氨基酸的变异。结论：指状青霉对原核细胞和真核细胞，无论在体内或体外均有致突变活性。

Study on the mutagenicity of Penicillium digitatum

Objective: We studied the mutagenicity of the extract from Penicillium digitatum. Methods: Revere mutation as-say, micronucleus test,unscheduled DNA syntnesis (UDS) in the primary cells of the lung and liver of rat and E. coli K12 in-fA gene mutation assay(the sequences of mutated infA gene were detemined and then analyzed by computer). Results: ①Theextract induced the reverse mutation of E. coli ND-160 strain;②The extract significantly did the increase of micronucleus rateof mouse bone marrow PCE(P＜0.01);③The extract could induce UDS in the primary cells of the lung and liver of rat,andUDS was particularly obvious in the primary cells of lung ( P＜0.01 ); ④Five points mutation in nucleotide of infA gene wereinduced by the extract and one of them led to amino acid variation(Lys→Val). Conclusion: Penicillium digitum could causegene mutation.
