IL-18对SLE患者外周血淋巴细胞凋亡及相关基因表达的研究

摘    要:本文研究SLE患者外周血淋巴细胞 (PBL )在IL 18刺激下细胞凋亡及凋亡相关基因表达情况。采用AnnexinV联合PI染色定量法及免疫荧光染色法。结果显示在IL 18刺激培养作用下 ,活动期SLE患者PBL凋亡发生率明显增高 (P <0 0 1) ,而静止期则无明显区别 (P >0 0 5 ) ,疾病活动性与PBL凋亡率呈正相关 (P <0 0 1)。Bcl 2表达活动期显著性升高 (P <0 0 5 ) ,静止期无明显著性差异 (P >0 0 5 )。Fas表达活动期显著升高 (P <0 0 0 1,静止期明显升高 (P <0 0 5 )。IL 18可引起SLE患者PBL凋亡率的增高 ,并导致调节紊乱 ,故在SLE发病机制中起作用

Abstract:The apoptosis and the expression of the apoptotic genes in PBL of patients with SLE stimulated by IL 18 were studied in 18 active SLE and 31 inactive cases by using annexin V united PI staining and immunofluorescence staining method It was found that the percentage of apoptosis in PBL of active cases stimulated by IL 18 was significantly higher than those of controls There was significant correlation between apoptotic percentage from SLE patients stimulated by IL 18 and the activities of disease Expression of Bcl 2 and Fas significantly elevated in active cases but showed no changes in inactive cases It concludes that IL 18 can induce increase of apoptotic percentage in patients with SLE,inducing disturbance of immunoregulation,and might play some role in the pathogenesis of SLE
