不同转移潜能人肺癌细胞系KAI1基因的表达

目的:探讨KAI1基因与人肺癌细胞系转移潜能的关系.方法:利用逆转录-PCR和Northern blot杂交法分析不同转移潜能人肺癌细胞系中KAI1 mRNA的表达,并用聚合酶链反应-单链构象多态性分析(PCR-SSCP)检测与突变的关系.结果:在高转移潜能细胞系PG中KAI1 mRNA表达降低,在无转移潜能的PAa细胞中KAI1的表达量比PG细胞高10倍以上(积分光密度比值分别为0.7313和0.0549).PCR-SSCP结果显示在PG细胞KAI1基因第7外显子出现异常改变.结论:肺癌细胞的转移潜能可能与KAI1基因表达量有关,KAI1基因的低表达可能由基因突变所致.

Expressions of KAI1 gene in human lung cancer cells with different metastatic potentials

Aim: To study the relation between KAI1 gene and metastatic potentials of human lung cancer cells. Methods: Expressions of KAI1 in human lung cancer cell lines with different metastasis potentials were detected by RT PCR and Northern blot hybridization, and the mutation were examined by PCR SSCP analysis. Results: In highly metastatic lung cancer cell line PG, expression of KAI1 mRNA was decreased and expression level in PAa, a lung cancer cell line with no metastasis potential was ten times higher than that in PG(integral light density values were 0.731 3 and 0.054 9 respectively). PCR SSCP analysis showed the exon 7 of KAI1 gene in PG appeared abnormal. Conclusion: Enhanced metastatic potential in human lung cancer cells may be associated with KAI1 expression. The decreased expression may be induced by mutation of KAI1 gene.
