脱蛋白液对异种骨移植替代物生物学特性影响

摘    要:目的研究不同脱蛋白时间对异种骨移植替代物细胞毒性、生物相容性和体内降解性的影响。方法采用脱蛋白液分别浸泡牛松质骨0、24、36、48和60h,通过与细胞共培养观察异种骨对骨髓基质细胞的细胞毒性,通过异种动物体内埋置观察其生物相容性和降解性。结果脱蛋白液处理48h和60h组细胞相对增殖率与空白对照组间无显著性差异(P〉0.05),细胞相容性最好,细胞毒性分级为0级;扫描电镜和组织学观察结果显示脱蛋白液处理48h组早期(4周内)即出现细胞长入软组织,具有良好的生物相容性;X线片、扫描电镜、能谱分析和显微CT观察显示48h组支架结构降解性明显优于其他组(P〈0.05),12周后48h组支架几乎完全降解。结论脱蛋白液处理48h后,异种骨移植替代物的细胞毒性最低,生物相容性和降解性达到最佳状态。
