凉血消沱汤对角质形成细胞凋亡的影响

目的研究自拟方剂凉血消疕汤对角质形成细胞株HaCaT的增殖凋亡及细胞周期的影响作用。方法用MTT法和Annexin-V/PI双标流式细胞术分别检测自拟方剂凉血消疕汤的水提取液对HaCaT细胞株增殖、凋亡的影响;PI单标流式细胞术测该方的水提取液对HaCaT细胞株细胞DNA分布的影响。结果自拟方剂凉血消疕汤对角质形成细胞株HaCaT具有明显的增殖抑制作用,其抑制增殖作用呈量效关系。凉血消疕汤作用48h可诱导HaCaT细胞凋亡,10mg/mL作用48h时HaCaT细胞凋亡率达(31.77±5.88)%。凉血消疕汤处理组HaCaT细胞株细胞周期发生变化,细胞周期G_1期增加,S期缩短。结论自拟方剂凉血消疕汤可通过调控细胞周期进程、诱导细胞凋亡而有效抑制HaCaT细胞株细胞增殖。

Effect of Decoction of Cooling Blood on Hacat Cell Apoptosis in Vitro

Objective To investigate the decoction of cooling blood on HaCaT cell growth and apoptosis.Methods The effect of decoction of cooling blood on the growth of HaCaT through MTT assay,the apoptosis of HaCaT by combined Annexin-V protein iodide staining,the cell cycle of HaCaT were studied by a propidium iodide method.Results Decoction of cooling blood inhibited the HaCaT cells in a time-manner,with 0.4～10mg/ mL.After 48h induced apoptosis of HaCaT cells,and the apoptosis rate reached(31.77±5.88 )%at concentration of 10mg/mL.Flow cytometric analyses showed that HaCaT cells treated with decoction of cooling blood were accumulated in G_1 phase in the cell cycle,the ratio of cells in and S phase was decreased at 48h treatment. Conclusion Decoction of cooling blood is able to inhibit the proliferation of HaCaT cells by regulating the cell cycle and inducing the cell apoptosis.
