肺纤维化大鼠模型肺组织病理超微结构观察及定量分析

摘    要:目的:观察以气管内灌注博来霉素方法建立的肺纤维化大鼠模型肺组织的病理演变规律,建立定量评价病变程度的方法.方法:30只大鼠随机分成5组:正常组,第7天模型组,第14天模型组,第21天模型组,第28天组,按时取肺组织进行电镜观察,以肺泡间隔面积和肺泡间隔灰度为定量分析指标,行光镜下的图像分析.结果:电镜观察显示肺泡间隔中胶原沉积随时间增多,与低倍镜下形态定量分析对比观察,肺泡间隔面积随时间增大,肺泡间隔灰度随时间减小.从第14天起与正常组比较差异有统计学意义.结论:低倍镜条件下肺泡间隔面积和肺泡间隔灰度基本能反映间隔中胶原沉积的程度,可以作为病变程度的一个主要评价指标.

Abstract:Objective: To observe the pathologic evolvement rule of the pulmonary fibrosis in rats model which was established with the method of intratracheal injection of bleomycin,and find the way for quantitative estimating of the degree of the pathological changes.Methods: Divided the 30 rates into 5 groups as: the control group,the 7th day group,the 14th day group,the 21st day group,the 28th day group. Took the lung tissue for electron microscope observation according to the time. Made the image analysis in microscope with the average and greyscale of the alveolar interval as the quantitative analysis index. Results: The electron microscope observation showed the collagen sediment in the alveolar interval increases with time.Contrasting with the morphology quantitative analysis in the low multiple microscope,the average of the alveolar interval increases with time and the greyscale of the alveolar interval decreased with time.There was significant difference from the 14th day in contrast with the control group. Conclusion: The avreage and greyscale of the alveolar interval can basically reflect the degree of the collagen sediment in the alveolar interval under the condition of the low multiple microscope.It can be used as an important evaluation index of the degree of the pathological changes.
