摘    要:目的 分析胰腺癌细胞和组织的基因不稳定性。方法 应用RAPD方法7种引物扩增4株胰腺癌细胞株BxPC-3、AsPC-1、SW1990和PaTu8988,3例手术切除胰腺癌及其癌旁组织,1例正常胰腺组织的DNA片段。结果 PaTu8988与SW1990扩增的DNA片段类型基本一致,但与BxPC-3、AsPC-1的类型不同;胰腺癌和癌旁组织扩增的DNA类型多数相同,少数存在差异,但与正常胰腺组织及胰腺癌细胞株不同。结论 胰腺癌组织中存在着基因不稳定性,这是造成肿瘤异质性的基础。

Abstract:Objective To analyze genomic instability in pancreatic cancer cell lines and tissues. Methods Genomic instability of pancreatic cancer cell lines BxPC-3,AsPC-1,PaTu8988,SW1990,three pairs of pancreatic cancer and noncancerous tissues,and one normal pancreatic tissue was detected by random amplified polymorphic DNA analysis using seven primers. Results Amplified DNA patterns from PaTu8988 were basically the same as those from SW1990,and differed from those of BxPC-3 and AsPc-1. Amplified DNA patterns of most pancreatic cancer tissues were the same as those of adjacent noncancerous tissues and differed from those of normal pancreatic tissue and pancreatic cancer cell lines. Conclusions There was genomic instability in pancreatic cancer tissues,resulting in heterogenecoty and heterogenecity of tumors.
