摘    要:目的建立一种新的简便、快速FTA法检测水产品中吸虫囊蚴。方法分别用FTA法和试剂盒法提取DNA,用定量PCR检测吸虫ITS2基因,区分淡水鱼中华支睾吸虫囊蚴、东方次睾吸虫囊蚴和溪蟹中并殖吸虫囊蚴。结果用FTA法能检测到每g水产品中1个吸虫囊蚴,定量PCR技术能区分水产品中3种不同的囊蚴,FTA法和试剂盒法具有相同的检测效果。结论 FTA法能快速检测水产品中吸虫囊蚴,具有简便、快速且敏感性高的优点,适宜海关防疫检验。

Abstract:In order to establish a new,simple and rapid FTA method for detection the metacercariae in aquatic products,FTA and kit methods were used in DNA extraction.ITS2 sequence has been amplifed to distinguish the metacercariae of Clonorchis sinensis,Metorchis orientalis and Paragonimus westermani in freshwater fishes and crabs in China.The result showed that one metacercariae per gram of fish was detected by FTA,and quantitative PCR technology could distinguish three kinds of metacercariae in aquatic products.Boeh FTA method and kit method have the same effect in the detection.It's suggested that FTA has the advantage of convenience,rapidity and high sensitivity in detection of metacercariae in aquatic products,and could be used in customs quarantine inspection.
