南昌市鼠类汉坦病毒检测及基因分型

摘    要:摘要:目的 了解从江西省南昌市捕捉的鼠类汉坦病毒感染情况,分析汉坦病毒基因型别和基因特征。方法 捕鼠取鼠肺,冷冻切片后运用直接免疫荧光法检测鼠肺中汉坦病毒抗原,提取抗原阳性鼠肺标本RNA,采用特异性引物对部分M基因片段进行基因扩增,对扩增产物进行序列测定和基因分析。结果 共捕获鼠类130只,褐家鼠(99,76.15%)、小家鼠(13,10.00%)、黄胸鼠(18,13.85%),其中18份汉坦病毒免疫荧光阳性,鼠类汉坦病毒携带率为13.85%,不同地区间汉坦病毒携带率差异有统计学意义(χ2=17.89,P=0.00129<0.05),青山湖区最高(26.67%);对所有的免疫荧光抗原阳性鼠肺标本进行核酸检测、序列测定和分析,结果发现所有标本汉坦病毒均属汉城型S4亚型,毒株之间核苷酸同源性986%~100.0%。结论 南昌市鼠间流行的汉坦病毒为汉城型,S4亚型。

Abstract:Abstract:Objective The study aimed to assess the status of hantavirus infection in rats captured in Nanchang City of Jiangxi Province and analyze the genotype and genomic features of hantavirus. Methods Rat lungs were obtained from captured rats and were subjected to frozen sectioning. Hantavirus antigen in the rat lungs was detected by direct immunofluorescence. RNA in antigen-positive rat lung specimens was extracted, and primers specific to parts of the M gene segment were used for gene amplification. The amplification products were sequenced and analyzed. Results A total of 130 rats were captured, including Rattus norvegicus (99, 76.15%), Mus musculus (13, 10.00%), and Rattus flavipectus (18, 13.85%). 18 of the specimens were tested positive of hantaviruse by immunofluorescence (13.85%); differences in positive rate of hantavirus were statistically significant among rats captures in different areas (χ2=17.89, P=0.00129<0.05), with the Qingshan Lake area having the highest positive rate (26.67%). All antigen-positive rat lung specimens as tested by immunofluorescence were subjected to nucleic acid test, gene sequencing, and analysis. The results showed that genotype of hantavirus in all the specimens was Seoul-S4, and nucleotide homology between viral strains was 98.6%-100.0%.Conclusion The hantavirus prevalent among rats in Nanchang City is Seoul-S4 hantavirus.
