大剂量糖皮质激素上调血管内皮细胞MR表达的意义

摘    要:目的 观察UPS和Dex对人脐静脉内皮细胞盐皮质激素受体(MR)的表达情况,以探讨糖皮质激素作用的新机制.方法 利用内毒素脂多糖LPS“致伤”血管内皮细胞,用地塞米松进行“治疗”,然后通过RT-PCR和免疫组化的方法检测血管内皮细胞盐皮质激素受体(MR)的表达情况.结果 内皮细胞本身能明显表达MRmRNA,运用不同浓度Dex和100 ng/mlLPS刺激后,可使人脐静脉内皮细胞内MRmRNA表达明显下降,应用大剂量(10-6mol/L)的Dex刺激细胞3h,MRmRNA的表达无明显变化,6h开始有升高,12 h达高峰,24 h回升近刺激前水平.免疫组化的结果与RT-PCR一致.结论 大剂量的DEX( 10-6mol/l)可上调MR的表达,GC引起GR和MR表达的不同变化可能是机体对损伤应激的一种调节机制,表明糖皮质激素作用存在新的机制.

Abstract:Objective To explore the new mechanism of glucocorticoid by observing the expression of glucocorticoid receptor(GR) and mineralocorticoid receptor(MR) with LPS and Dex in human umbilical vein endothelial cells.Methods lipopolysaccharide LPS "injured" endothelial cells with dexamethasone for "treatment",and then detected the expression of mineralocorticoid receptor(MR) in the endothelial cells by RT-PCR and immunohistochemistry.Results Using different concentrations of Dex and 100 ng/ml LPS stimulation,human umbilical vein endothelial cells MRmRNA expression decreased,and using high dose(10-6 mol/L) of Dex to stimulate cell 3 h,MRmRNA had no significant changes in expression.The expression of MRmRNA began to increase at 6h,peaked at 12 h,and then rebound near the level before stimulation at 24 h.Immunohistochemistry results were consistent with the RT-PCR.Conclusion Large dose of DEX(10-6 mol/L) up-regulated the expression of MR.GC induced the different expression of GR and MR to stress of injury of the body,which may be a regulatory mechanism and may even indicate a new mechanism of glucocorticoid.
