摘 要: | 基质金属蛋白酶(MMPs)是一种能降解各种细胞外基质(ECM)成分的蛋白水解酶家族,内源性金属蛋白酶类组织抑制剂(TIMPs)可以减少MMPs对ECM蛋白的过度降解。对内源性TIMPs的表达进行基因操作或者通过人工合成的药物抑制剂使MMPs的表达下调可能对减少MMPs介导的血管功能障碍和血管壁的过度损害等血管疾病发挥益处。了解其作用机制有助于预防和治疗相关疾病。

Matrix metalloproteinase and the prevention and treatment of restenosis after vascular injury |

Abstract: | Matrix metalloproteinases (MMPs) are a family of proteolytic enzymes that degrade various components of the extracellular matrix (ECM). Endogenous tissue inhibitors of metalloproteinases (TIMPs) reduce ex- cessive proteolytic ECM degradation by MMPs. Down regulation of MMPs using genetic manipulations of endogenous TIMPs,or synthetic pharmacological inhibitors could be beneficial in reducing the MMP-mediated vascular d- ysfunction and the progressive vessel wall damage associated with va- scular disease. To understand their mechanism of action will contribute to the prevention and treatment of related diseases. |
