脂联素对大鼠正畸牙移动中牙周组织RANKL表达的影响

摘    要:目的:探讨局部注射脂联素对大鼠正畸牙移动距离与牙周组织骨改建的影响.方法:30只SD大鼠双侧上颌建立正畸牙移动模型,随机分为2组,加力第1天起在实验组正畸牙颊侧牙龈黏膜下局部注射脂联素,对照组注射1%磷酸缓冲液(PBS),每2天1次.两组大鼠加力后分别于第1,3,7,10,14天处死.测量牙移动距离,用HE染色观察被移动磨牙牙周组织形态学变化,应用免疫组织化学染色法对牙周组织中RANKL进行半定量分析.结果:实验组在第7,10,14天牙齿移动距离和RANKL阳性表达均比对照组明显减小,差异有统计学意义(P<0.01).结论:脂联素抑制牙周组织RANKL的表达,参与了正畸牙移动过程中的牙周组织骨改建,外源性脂联素可以减少牙齿移动距离.

Abstract:Objective:To investigate the effects of local administration of adiponectin solution on the orthodontic tooth movement and periodontal ligament.Methods:An experimental model of orthodontic tooth movement was established in rats.Thirty SD rats were randomly divided into 2 groups.The experimental group was local injected with adiponectin in the buccal submucosal area of the molar,whereas the control group was injected with PBS.The injection was applied every two day.The rats were sacrificed on days 1,5,7,10 and 14.The distance of tooth movement was measured after sacrificed,sections were HE-stained for observing the activities of periodontal tissues rebuilding,immunohistochemical staining was used for analysing of RANKL expression in the periodontal tissues.Results:The distance of tooth movement and RANKL positive expression was lower in the experimental group than in control group on days 7,10 and 14 (P 〈0.01).Conclusion:Adiponectin could inhibit the expression of RANKL in periodontal tissue,reduce the distance of tooth movement.
