L苯丙氨酸抑制自发性高血压大鼠心肌成纤维细胞增殖

摘    要:目的旨在观察L-苯丙氨酸对自发性高血压大鼠心肌成纤维细胞的增殖作用的影响。方法取1天龄自发性高血压大鼠心脏组织培养心肌成纤维细胞,分别以不同浓度的L-苯丙氨酸、酪氨酸和组氨酸干预一定时间后,利用3H-胸腺嘧啶掺入技术和细胞计数测定各不同浓度的氨基酸对心肌成纤维细胞DNA合成和细胞增殖数目的影响。结果随着L-苯丙氨酸浓度的增加,心肌成纤维细胞的DNA合成量减少,细胞增殖数目减低。而L-酪氨酸和组氨酸随着浓度的增加对心肌成纤维细胞DNA的合成量和细胞增殖数目无影响。结论L-苯丙氨酸可以特异性地抑制SHR心肌成纤维细胞的增殖。

Abstract:Objective To observe the effect of LPhenylalanine on proliferation of myocardial fibroblast in spontaneosly hypertensive rat. Methods Amino acids in different concerntration were added to myocardial fibroblast cultured from 1 dayold spontaneously hypertensive rat for a defined time. The3Hthymidine incorporation was applied to reflect the fibroblasts proliferation activity with counts of3Hthymidine per minute divided by the number of fibroblasts. Result The DNA synthesis/cell count were decreased with the Phenylalanine in a dose dependet manner, but not with tyrosine and histidine. Conclusion LPhenylalanine inhibited the proliferation of myocardial fibroblast in spontaneously hypertensive rat, which may explain the proven cases of phenylalanine preventing blood pressure rise and myocardial hypertrophy in spontaneously hypertensive rat.
