盐酸戊乙奎醚对大鼠急性脊髓损伤修复的作用

摘    要:目的 观察盐酸戊乙李醚对大鼠急性脊髓损伤后脊髓损伤修复的作用.方法 选用健康成年雄性SD大鼠90只分为3组,A组:盐酸戊乙奎醚治疗组(n=40),从造模后1 h开始,经腹腔注射盐酸戊乙奎醚每天3 mg/kg,卣至取材;B组:单纯损伤组(n=40),单纯损伤组同法给予等量生理盐水;C组:正常对照组(n=10),不作任何处理.A组、B组大鼠采用改良Allen'S重物坠落法在T10段制作急性脊髓损伤模型.各组取材前24 h腹腔注射溴脱氧尿嘧啶核苷(BrdU)溶液,对距离损伤中心5 mm处的脊髓进行BrdU、nestin阳性细胞数检测.结果 C组脊髓中央管周围及外膜可见少量BrdU阳性细胞,几乎看不到nestin阳性细胞.B组造模后24 h即可见大量BrdU阳性细胞,1周达到高峰,2周后开始明显减少,4周时仅见少量BrdU阳性细胞.与B组比较,24 h时A组BrdU阳性细胞差异无统计学意义,1周时BrdU阳性细胞数明显增多(P<0.05),至2周时仍处于较高水平,4周时仍有大量BrdU阳性细胞表达.B组造模后7 d nestin阳性细胞增多,14 d达到高峰,28 d时可见极少量nestin瞄H性细胞.与B组比较,7 d时A组nestin阳性细胞差异无统计学意义,14 d时nestin阳性细胞数明显增多,至28 d时仍处于较高水平(P<0.05).结论 盐酸戊乙奎醚能够促进急性脊髓损伤大鼠损伤区BrdU及nestin的表达,提示有促进神经干细胞增殖的能力,增强脊髓损伤后自体的修复功能.

Abstract:Objective To explore the effect of Penehyclidine Hydrochloride Injection on the proliferation of spinal cord stem cells in the rats with spinal cord injury.Methods Ninty Sprague-Dawley rats were randomized into three groups:Group A (Penehyclidine Hydroehloride treated group,n=40),Group B(simple injury group,n=40)and Group C(normal control group,n=10)(1,3,7,14 and 28 days),rat models of acute spinal cord injury at TIO segment were made using Allen'S method from 1 hour following model induction.Rats in the penehyctidine hydrochloride group received 3mg/kg/d intraperitoneal injected penehyelidine hydroehloride.Samples were directly obtained.In the simple injury group,rats received an equal volume of saline.Rats in the normal control group were left untreated.24 hours before samples were obtained,rats in each group were intraperithoneally injected with bmmodeoxyuridine (BrdU) solution for labeling.once every 8 hours and 3 times in total.Spinal Cord at 5 mm from the center of damage region received BrdU nestin immunohistochemistry.BrdU-positive cell number and area of nestin-positive expression were counted.Results There were a few BrdU-positive cells and less nestin-positive cells in the center canal and adventi tia of spinal cord in the normal control group.Many BrdU-positive cells were found in the injury area in the simple injury group 24 hours after model induction,which reached the peak at 1 week,and declined gradually at 2 weeks.Just a few BrdU-positive cells existed at 4 weeks.Compared with the simple injury group,there was no difference in BrdU-positive cell at 24 hours following model induction in the penehyelidine hydrochlofide group.There were more BrdU-positive cells in the center canal and advenfitia of spinal cord at 1 week(P<0.05)and still high at 2 weeks and 4 weeks.Many nestin-positive eels were found in the injury area in the simple injury group 7 days after model induction,which reached the peak at 2 week,just a few nestin-positive cells existed at 4 weeks.Compared wilh the simple injury group.there was no difference in nestin-positive ceils at 7 days following model induction in the penehyclidine hydrochloride group.There were more nestin-positive cells in the center canal and adventitia of spinal cord at 2 week and still high at 4 weeks(P<0.05).Conclusion Spinal cord injury(SCI)stimulates the proliferation of inherent neural stem cells.1his indicated that ponehyelidine hydrochloride could significantly promote proliferation ofendogenous neural stem cells,therefore encourage the recovery of neural function.
