夏天无注射液对大鼠脑缺血再灌损伤及ICAM-1 mRNA表达的影响

摘    要:目的探讨夏天无注射液(Injection Coryadlis Decumbens Pers,ICDP)对大鼠脑缺血再灌的影响及其作用机制。方法制备大鼠大脑中动脉阻塞2h再灌注24h模型,应用氯化三苯四氮唑(triphenyltetrazolium chloride,TTC)染色测量脑组织梗死面积;逆转录—定量多聚酶链反应(RT-real time PCR)方法,检测ICDP对大鼠脑缺血再灌后海马细胞间粘附分子-(1 intercelluar adhesion molecule-1,ICAM-1)mRNA的水平。结果与模型组比较,大鼠脑缺血再灌注损伤后ICDP 0.05ml/kg、0.025 ml/kg剂量组脑梗死面积明显减少(P〈0.01或P〈0.05);ICDP 0.01ml/kg组脑梗死面积与模型组比较无显著差异;ICAM-1 mRNA在各组中表达无明显差异。结论夏天无注射液对脑缺血再灌注损伤具有明显保护作用,其作用机制可能不涉及ICAM-1 mRNA的水平变化。

Abstract:Objective To investigate the protective effect and mechanism of ICDP on focal cerebral ischemia reperfusion injury in rats.Methods The operation was performed to induce the middle cerebral artery occlusion(MCAO) model in rats with the method of embolic thread.The middle cerebral artery blood flow was blocked for 2 hours,and then reperfused twenty-four hours later.Brain tissue was stained by TTC.The infarct size was measured by wet weight method.The level of ICAM-1 mRNA expression was measured by RT-real time PCR.Results The infarct size and the proportion of infarct size(PI) were significantly decreased in ICDP groups(0.05 ml/kg and 0.025 ml/kg) compare with in the model group.But the level of ICAM-1 mRNA expression were not differences(P>0.05) in every groups.Conclusions ICDP has significant protective effect on focal cerebral ischemia reperfusion injury,but the mechanism may not be related in ICAM-1 mRNA expression..
