摘    要:目的:检测NOD2在舌鳞癌中的表达特征及功能变化。方法:采用免疫组织化学、RT-qPCR检测人舌鳞癌和正常舌黏膜组织中NOD2的表达;采用免疫荧光、RT-qPCR检测NOD2活化对人舌鳞癌细胞株Tca8113、正常舌黏膜上皮细胞分泌hBD-2的影响。应用SPSS 13.0软件包对各组mRNA的表达进行t检验及多个样本均数的方差分析。结果:①NOD2蛋白表达于细胞质,舌鳞癌组织中NOD2的蛋白表达率为64.0%(16/25),显著低于正常舌黏膜组织的表达率84.0%(21/25,P<0.05)。舌鳞癌组织NOD2 mRNA表达量为正常舌黏膜组织的(0.521±0.080)倍(P<0.05)。②hBD-2表达于细胞质,Tca8113细胞中hBD-2为低表达和阴性表达,MDP处理6 h、12 h后,Tca8113细胞中hBD-2mRNA的表达量较处理前显著上调(1.770±1.202)、(68.277±48.946)倍(P<0.05)。结论:舌鳞癌组织中NOD2 mRNA和蛋白表达显著低于正常舌黏膜组织,NOD2激动剂MDP能上调Tca8113细胞中hBD-2 mRNA和蛋白的表达。推测在舌黏膜中NOD2能调节hBD-2表达,可能影响舌鳞癌的免疫功能,与舌鳞癌进展相关。

Abstract:PURPOSE: This study aimed to investigate the expression and functional changes of NOD2 in tongue squamous cell carcinoma (TSCC). METHODS: Immunohistochemical staining and RT-qPCR were employed to evaluate NOD2 expression in TSCC and non-cancer tongue epithelial tissues, hBD-2 production was investigated by immunofluorescenee staining and RT-qPCR. Student's t test was used to assess the statistical significance of mRNA expression between the two groups, and two-way analysis of variance was used to determine the differences among different groups using SPSS 13.0 software package. RESULTS: ①NOD2 was expressed in the cell plasma. NOD2 expression was significantly lower in TSCC tissues (64.0%, 16/25) than in non-cancer tongue epithelial tissues (84.0%, 21/ 25) (P〈0.05). NOD2 messenger RNA expression in TSCC tissues was (0.521±0.080) times of that in non-cancer tongue epithelial tissues(P〈0.05). ②hBD-2 was expressed in the cell plasma. When MDP was added to TcaS113 cells for 6,12 h, hBD-2 expression was significantly upregulated (1.770±1.202 and 68.277±48.946 times, P〈0.05). CONCLUSION: NOD2 expression was significantly lower in TSCC, hBD-2 expression was markedly upregulated when NOD2 was activated by MDP. This suggests that NOD2 adjust hBD-2 expression in the tongue mucosa, and affect the immune function of TSCC, as well as the progression of TSCC.
